Microglia are implicated in the pathophysiology of several neurodegenerative disorders, including Alzheimer's disease. While the role of microglia and peripheral macrophages in regulating amyloid beta pathology has been well characterized, the impact of these distinct cell subsets on tau pathology remains poorly understood. We and others have recently demonstrated that monocytes can engraft the brain and give rise to long-lived parenchymal macrophages, even under nonpathological conditions. We undertook the current study to investigate the regulation of tau pathology by microglia and peripheral macrophages using hTau transgenic mice, which do not exhibit microglial activation/pathology or macrophage engraftment. To assess the direct impact of microglia on tau pathology we developed a protocol for long-term microglial depletion in Cx3cr1(CreER)R26(DTA) mice and crossed them with hTau mice. We then depleted microglia up to 3 months in both young and old mice, but no net change in forebrain soluble oligomeric tau or total or phosphorylated levels of aggregated tau was recorded. To investigate the consequence of peripherally-derived parenchymal macrophages on tau aggregation we partially repopulated the hTau microglial pool with peripheral macrophages, but this also did not affect levels of tau oligomers or insoluble aggregates. Our study questions the direct involvement of microglia or peripheral macrophages in the development of tau pathology in the hTau model.

Osman, Dong Liang, Wei Han, Kristina Gemzell-Danielsson, Christer Betsholtz, Xing-Mei Zhang, Changlian Zhu, Martin Enge, Bertrand Joseph, Robert A. Harris, Klas Blomgren

Summary: A subpopulation of microglia lacking genetic modification and showing high CX3CR1 expression was identified in Cx3cr1(CreER-Eyfp/wt) mouse strain. This subpopulation escaped depletion and proliferated extensively after microglial depletion, eventually occupying a large proportion of microglial pool. Additionally, it was demonstrated that CX3CL1-CX3CR1 signaling regulates microglial repopulation.

Jing-xian Sun, Ke-ying Zhu, Yu-meng Wang, Dan-jie Wang, Mi-zhen Zhang, Heela Sarlus, Irene Benito-Cuesta, Xiao-qiang Zhao, Zao-feng Zou, Qing-yang Zhong, Yi Feng, Shuai Wu, Yan-qing Wang, Robert A. Harris, Jun Wang

Summary: The role of TRPV1 in demyelinating diseases has been investigated, and it has been found to be an important regulator in the context of demyelination. TRPV1 deficiency exacerbates motor dysfunction and demyelination, while TRPV1 agonist CAP improves behavioral performance and facilitates remyelination. The activation of TRPV1 enhances microglial function and promotes myelin debris clearance.

Microglial aryl hydrocarbon receptor enhances phagocytic function via SYK and promotes remyelination in the cuprizone mouse model of demyelination

Yumeng Wang, Jingxian Sun, Keying Zhu, Danjie Wang, Xiaoqiang Zhao, Hongyu Zhang, Shuai Wu, Yanqing Wang, Jun Wang

Summary: Multiple sclerosis (MS) is an inflammatory-mediated demyelinating disease of the central nervous system (CNS). Microglia with aryl hydrocarbon receptor (AhR) play a crucial role in efficient remyelination and clearance of myelin debris. AhR deficiency in microglia impairs phagocytic function, exacerbates CNS inflammation, and inhibits effective remyelination. This study highlights the therapeutic potential of AhR in demyelinating diseases.

Study on Mechanical Property of Coal-Rock Combination Under Different Unloading Confining Pressure Rate

Shanchao Hu, Xuedong Zhou, Wenkai Ru, Jinming Han, Shihao Guo, Chenxi Zhang, Lei Yang

Summary: This study conducted unloading confining pressure tests on coal-rock combinations under three different pressure rates. The stress-strain curves of the combinations were observed, and their failure, deformation, and strength characteristics were analyzed. The results showed that the combinations exhibited obvious volume expansion due to simple shear failure of the coal during unloading of the confining pressure. The rock part started to damage with increasing confining pressure value and unloading rate. The fracture angle was proportional to the initial confining pressure and inversely proportional to the unloading confining pressure rate, while the fragmentation and intensity degree varied significantly under different test conditions.
