F2-isoprostanes in Fish mucus: A new, non-invasive method for analyzing a biomarker of oxidative stress

F-2-isoprostanes (F-2-isoPs) are a reliable biomarker class for oxidative stress in vivo in animals. These compounds are traditionally measured in matrices like liver and plasma, however social and environmental pressures warrant the development of non-lethal and non-invasive methods to assess animal health. Therefore, this study aimed to develop a high-performance liquid chromatography tandem mass spectrometry (HPLC-ESI-MS/MS) method to separate and detect F-2-isoPs in fish mucus. The method was developed and validated for four native F-2-isoP isomers using Northern pike mucus (Esox lucius). Linearity was observed between 5 and 1000 pg/mu L. The limits of detection of the four F-2-isoP isomers ranged from 0.63 to 2.0 ng/g. Recoveries ranged from 78 to 95%, and matrix effects were small (<10%). The between-day and within-day repeatability for all target analytes was lower than 20% RSD. Endogenous F-2-isoPs were measured in the pike mucus (5.3-28.8 ng/g). A preliminary study of baseline F-2-isoP concentrations in lake trout (Salvelinus namaycush) captured from five lakes at the IISD-Experimental Lakes Area in Northwestern Ontario, Canada, was also conducted to test the interspecies applicability of the method. Endogenous F-2-isoPs were quantified in lake trout (63-132 ng/g). Lake trout samples displayed large variability within and between the different lakes, which suggests sampling methods may require adjustment for this species. This work developed a sensitive analytical method for measuring F-2-isoPs in fish mucus, however several further studies are required to determine its ability to accurately measure oxidative stress in fish species. New insight on occurrence of liquid crystal monomers: A class of emerging e-waste pollutants in municipal landfill leachate

Summary: A robust method was developed to determine 38 target liquid crystal monomers in landfill leachate. 23 and 20 LCMs were detected in samples collected from municipal landfills in Hong Kong and Shenzhen, respectively, suggesting that landfill leachate may be a potential sink of LCMs emitted from e-waste. Further research is needed to investigate the potential migration of LCMs from landfill leachate and their ecotoxicological risk in aquatic environments.

Metabolic transformation of environmentally-relevant brominated flame retardants in Fauna: A review

Summary: In the flame retardant industry, there has been a trend of using more highly brominated compounds as replacements for banned and regulated compounds. However, there is a lack of knowledge regarding the metabolism and transformation potential of these new compounds. Current research shows variations in experimental protocols and a lack of information on the fate and metabolic pathways of certain environmentally relevant flame retardants. More studies are needed to understand the bioaccumulation and fate of these compounds in exposed organisms.

Suspect Screening of Liquid Crystal Monomers (LCMs) in Sediment Using an Established Database Covering 1173 LCMs

Summary: This study reviewed existing literature on liquid crystal monomers (LCMs) and established a database containing 1173 LCMs. The physicochemical properties of these LCMs were calculated, and a suspect screening strategy tailored for LCMs was successfully developed. This strategy has the potential to be applied to other environmental matrices.

Suspect and nontarget screening of known and unknown organophosphate esters (OPEs) in soil samples

Summary: Ninety-five soil samples were analyzed to identify different OPEs and OPE-like substances. A total of 26 OPEs or OPE-like substances were identified, with BPABDP being the most frequently detected. The study also discovered four novel OPE structures, one of which shared a similar backbone structure with a well-known OPE, TDTBPP. This research provides the most comprehensive suite of OPEs in soil samples reported so far.

Dietary intake assessment of known and unknown organophosphate esters (OPEs) in foodstuffs via high-resolution mass spectrometry

Summary: An integrated screening strategy was applied to analyze OPEs in 107 foodstuffs collected from Nanjing City, China in 2020. The results showed that meat samples were the most severely contaminated. Additionally, suspect and non-target analysis identified other OPEs, providing important information for food safety assessment.

Tissue-specific accumulation, bioaccumulation, and depuration of liquid crystal monomers (LCMs) in adult zebrafish (Danio rerio)

Summary: This study investigated the bioaccumulation, tissue-specific accumulation, and depuration properties of liquid crystal monomers (LCMs) in adult zebrafish. The study found that the accumulation potential of LCMs varied among different functional groups, and LCMs with the cyan group were more likely to accumulate in zebrafish. The study also suggested that the bioaccumulation potential of some LCMs in aquatic environments might be overestimated by theoretical estimation techniques.

Target and suspect analysis of liquid crystal monomers in soil from different urban functional zones

Summary: Recent studies have found the widespread presence of liquid crystal monomers (LCMs) in various environments, but there is limited information about their contamination in soil. This study collected soil samples from different urban functional zones and found that LCMs are widely distributed in surface soil. The agricultural zone had the highest concentration of LCMs, followed by commercial, residential, industrial, and scenic zones. Correlations between different LCMs were also observed, suggesting similar commercial applications and sources.

Microbead-Beating Extraction of Polycyclic Aromatic Compounds from Seabird Plasma and Whole Blood

Summary: Seabirds are valuable bioindicators of environmental health, and we have developed a new method to extract polycyclic aromatic compounds (PACs) from seabird plasma and serum. The method involves automated microbead-beating homogenization and extraction, and has shown good accuracy and repeatability for extracting PACs from important biological matrices.

Tissue-specific uptake and distribution of liquid crystal monomers (LCMs) in mice

Summary: Increasing evidence suggests that liquid crystal monomers (LCMs) in liquid crystal displays can be released into the environment and detected in environmental matrices and human bodies. However, there is a lack of databases regarding their uptake and distribution in mammals. This study investigated the distribution and accumulation of four LCMs in mice tissues, revealing their preferential deposition in lipophilic tissues and the influence of physico-chemical properties on their distribution and accumulation. These findings have important implications for biomonitoring and risk assessment of LCMs.

Liquid Crystal Monomer: A Potential PPARγ Antagonist

Summary: Liquid crystal monomers (LCMs) are commonly used in liquid crystal display (LCD) industries, but their environmental occurrences and toxic effects have not been thoroughly studied. This study used in silico simulation, in vitro assays, and omics integration analysis to comprehensively investigate the toxicity and mechanism of LCMs. The results showed that certain LCMs significantly antagonized peroxisome proliferator-activated receptor gamma (PPARγ), leading to dysregulation of metabolome and transcriptome in HK2 cells. Dysregulation of fatty acid beta-oxidation, mediated through multiple pathways, and the activation of AMPK and ligand-dependent PPARγ antagonism were identified to play important roles. This study provides valuable insights for future chemical risk assessment.

Characterizing the obesogenic and fatty liver-inducing effects of Acetyl tributyl citrate (ATBC) plasticizer using both in vivo and in vitro models

Summary: The incidence of obesity and non-alcoholic fatty liver disease (NAFLD) has been increasing rapidly worldwide. Environmental factors such as plastic usage and chemical exposure have been suggested to be important contributors to the obesity epidemic. Acetyl tributyl citrate (ATBC), a commonly used non-phthalate plasticizer, has a high leakage rate from plastics, leading to increased exposure risk. This study aimed to evaluate the safety of subchronic exposure to environmentally-relevant concentrations of ATBC. Results showed that ATBC exposure led to weight gain, increased body fat content, larger adipocytes, and liver steatosis in mice. In vitro experiments also demonstrated increased intracellular lipid accumulation with ATBC treatment.

Plastic packaging-associated chemicals and their hazards-An overview of reviews

The study found that there are over 10,259 chemicals related to plastic packaging materials, and it discussed their structure, analysis methods, migration characteristics, and potential hazards.

Organophosphate esters (OPEs) in plastic food packaging: non-target recognition, and migration behavior assessment

Summary: An integrated screening method was optimized using UHPLC-HRMS to investigate the migration of OPEs from plastic food packaging. A total of 42 OPEs were identified, including seven new ones, in 106 samples collected in Nanjing, China. The study also revealed the oxidation products of AO626 in plastics, suggesting the importance of the oxidation of OPAs as an indirect source of OPEs. Migration of OPEs was observed in four simulated foods, with diverse OPEs detected at elevated concentrations in isooctane.

Determining Polycyclic Aromatic Compounds in Bird Feathers Using Pressurized Fluid Extraction

Summary: Bird feathers are increasingly used as a non-invasive biomonitoring tool for environmental monitoring. A validated method for measuring polycyclic aromatic compounds (PACs) in bird feathers using pressurized fluid extraction and gas chromatography-tandem mass spectrometry is presented. The method provides accurate and efficient extraction and quantitation of PACs, eliminating the need for time-consuming acid treatment.
