The MEK5-ERK5 Kinase Axis Controls Lipid Metabolism in Small-Cell Lung Cancer

Small-cell lung cancer (SCLC) is an aggressive form of lung cancer with dismal survival rates. While kinases often play key roles driving tumorigenesis, there are strikingly few kinases known to promote the development of SCLC. Here, we investigated the contribution of the MAPK module MEK5-ERK5 to SCLC growth. MEK5 and ERK5 were required for optimal survival and expansion of SCLC cell lines in vitro and in vivo. Transcriptomics analyses identified a role for the MEK5-ERK5 axis in the metabolism of SCLC cells, including lipid metabolism. In-depth lipidomics analyses showed that loss of MEK5/ERK5 perturbs several lipid metabolism pathways, including the mevalonate pathway that controls cholesterol synthesis. Notably, depletion ofMEK5/ERK5 sensitized SCLC cells to pharmacologic inhibition of the mevalonate pathway by statins. These data identify a new MEK5- ERK5-lipid metabolism axis that promotes the growth of SCLC. Significance: This study is the first to investigate MEK5 and ERK5 in SCLC, linking the activity of these two kinases to the control of cell survival and lipid metabolism.

Summary: Understanding the impact of genetic variants on molecular phenotypes is important in functional genomics. The EN-TEx resource provides 1,635 open-access datasets mapped to diploid genomes, revealing over 1 million allele-specific loci. Deep-learning models can predict allele-specific activity based on local nucleotide-sequence context, emphasizing the importance of transcription-factor-binding motifs sensitive to variants. EN-TEx also shows strong associations between allele-specific and GWAS loci, allowing for the transfer of known eQTLs to difficult-to-profile tissues. Overall, EN-TEx provides rich data and generalizable models for more accurate personal functional genomics.

Summary: There is a need to identify blood-borne factors that mediate tissue crosstalk during physical activity. This study used a cell-type-specific proteomic approach to map exercise training-regulated secretomes in mice, identifying over 200 previously unreported cell-type-secreted protein pairs. The study also found exercise-induced secretion of proteoforms of intracellular carboxylesterases from the liver, which had anti-obesity, anti-diabetic, and exercise performance-enhancing activities.

Summary: We developed an analysis pipeline called spatial metatranscriptome (SMT) that extracts microbial sequences from spatial transcriptomic (ST) data, assigns taxonomic labels, and generates a spatial microbial abundance matrix. The pipeline was applied to human and murine intestinal sections and validated with alternative assays. This work provides biological insights into host-microbe interactions at different spatial scales and demonstrates the feasibility of SMT analysis.

Summary: This study investigates the phenotype and molecular mechanisms of vasculogenic mimicry (VM) in small cell lung cancer (SCLC) using circulating tumor cell-derived explant (CDX) models and genetically engineered mouse models (GEMMs). The results show that VM vessels are present in CDX models, GEMMs, and SCLC patient biopsies, and perfused VM vessels support tumor growth. Only NOTCH-active non-NE cells are VM-competent and exhibit characteristics related to blood vessel development and extracellular matrix organization. On Matrigel, VM-primed non-NE cells remodel the extracellular matrix into hollow tubules through an integrin b1-dependent process.

Summary: Organ Mapping Antibody Panels are standardized antibody panels created by the community for multiplexed spatial imaging. This imaging technique allows detailed characterization of molecular and cellular organization in tissues. Although high-parameter data collection is now possible (>60 targets), constructing the necessary antibody panels requires expertise and capital. Organ Mapping Antibody Panels are community-validated resources that save time and money, enhance reproducibility, expedite discovery, and support the creation of a Human Reference Atlas.

Summary: Combining AKT inhibitor and SERD improved PFS in a clinical trial for HR+ breast cancer patients. However, CDK4/6 inhibitor treatment may reduce response to subsequent endocrine treatment. This study explored the impact of CDK4/6 inhibitor treatment on breast tumor cell function and response to fulvestrant and capivasertib.
