Simple production of hydrophobin-fused domain III of dengue envelope protein and induction of neutralizing antibodies against the homotypic serotype of dengue virus

Hydrophobin-fused domain III of dengue envelope proteins serotypes 1 and 2 were expressed in Rachiplusia nu larvae and purified by aqueous two-phase system. This biotechnological approach of hydrophobin-fused proteins, which allowed obtaining 97.7 mu g/larva of fusion protein DomIII serotype 1 and 61.4 mu g/larva of fusion protein DomIII serotype 2, represents an integrated strategy for simple production of recombinant antigens. Purified fusion proteins induced serotype-specific neutralizing antibodies without cross-reaction against other serotypes and arboviruses after mouse immunization. hydrophobin-fused domain III of dengue envelope protein could be a promising strategy for easy and low-cost production of components of a tetravalent sub-unit vaccine against dengue.

Summary: Helminth infections pose a significant global health concern with over one billion people affected, and despite efforts, there is no effective human vaccine. The current main drugs used to control helminths are facing increasing resistance in livestock, leading to a need for alternative treatments like plant extracts or probiotics. Probiotics, by definition, are live microorganisms that can benefit the host's health when consumed in adequate amounts, and they have been shown to have immunomodulatory effects that could help in controlling gastrointestinal helminth infections.

Lipoteichoic Acid from Lacticaseibacillus rhamnosus GG Modulates Dendritic Cells and T Cells in the Gut

Summary: Lipoteichoic acid (LTA) from different Gram-positive bacterial sources has varying immune effects. Lacticaseibacillus rhamnosus GG LTA (LGG-LTA) oral administration reduces UVB-induced immunosuppression and skin tumor development in mice. In this study, the immunomodulatory effects of LGG-LTA on dendritic cells (DC) and T cells were evaluated in vitro and in the gut-associated lymphoid tissue (GALT). The results showed that LGG-LTA activated DC and T cells, promoting Th1-biased immune response after oral LTA administration.

Summary: This study describes a novel ELISA for antibodies testing and a dot blot assay for antigen testing using Hcp100 protein and their polyclonal antibodies, respectively, to diagnose PDH. The results showed that both methods have high sensitivity and specificity for detecting PDH and can serve as valuable screening tools.

Summary: In this study, we conducted an in-depth investigation into the activity of amidinoquinoxaline N-oxides 1 against Gram-positive and Gram-negative anaerobic bacteria. We found that these compounds showed good to excellent antianaerobic activity, low cytotoxicity, and potential as oral drug candidates.
