Cytotoxic potential of selected medicinal plants in northeast Brazil

Background: Great biodiversity is a highlight of Brazilian flora. In contrast, the therapeutic potentialities of most species used in folk medicine remain unknown. Several of these species are commonly used to treat cancer. In this study, we investigated the cytotoxic activity of 18 plants from 16 families that are found in the northeast region of Brazil. Methods: The following species were studied: Byrsonima sericea DC. (Malpighiaceae), Cupania impressinervia Acev. Rodr. var. (revoluta) Radlk (Sapindaceae), Duranta repens Linn. (Verbenaceae), Helicostylis tomentosa (Poepp. & Endl) Rusby (Moraceae), Himatanthus bracteatus (A. DC.) Woodson (Apocynaceae), Ipomoea purga (Wender.) Hayne (Convolvulaceae), Ixora coccinea Linn. (Rubiaceae), Mabea piriri Aubl. (Euphorbiaceae), Miconia minutiflora (Melastomataceae), Momordica charantia L. (Cucurbitaceae), Ocotea glomerata (Nees) Mez (Lauraceae), Ocotea longifolia Kunth (Oreodaphne opifera Mart. Nees) (Lauraceae), Pavonia fruticosa (Mill.) Fawc. & Rendle (Malvaceae), Psychotria capitata Ruiz & Pav. (Rubiaceae), Schefflera morototoni (Aubl.) Maguire, Steyerm. & Frodin (Araliaceae), Solanum paludosum Moric. (Solanaceae), Xylopia frutescens Aubl. (Annonaceae) and Zanthoxylum rhoifolium Lam. (Rutaceae). Their dried leaves, stems, flowers or fruits were submitted to different solvent extractions, resulting in 55 extracts. After incubating for 72 h, the cytotoxicity of each extract was tested against tumor cell lines using the alamar blue assay. Results: The B. sericea, D. repens, H. bracteatus, I. purga, I. coccinea, M. piriri, O. longifolia and P. capitata extracts demonstrated the most potent cytotoxic activity. The chloroform soluble fractions of D. repens flowers and the hexane extract of I. coccinea flowers led to the isolation of quercetin and a mixture of alpha- and beta-amyrin, respectively, and quercetin showed moderate cytotoxic activity. Conclusion: The B. sericea, D. repens, H. bracteatus, I. purga, I. coccinea, M. piriri, O. longifolia and P. capitata plants were identified as having potent cytotoxic effects. Further investigations are required to determine the mechanisms of cytotoxicity exhibited and their in vivo activities. This work reinforces the need to understand the therapeutics potentialities of Brazilian medicinal plants.

Biological studies and chromatograms aided by chemometric analysis in evaluation of seasonality and extraction method of Croton grewioides extracts

Summary: This study investigated the effects of seasonal variability and extraction method on the biological activities of polar extracts from Croton grewioides. The results showed that seasonal changes only affected the chemical composition of certain extracts. Different extraction methods also led to variations in the chemical profiles of the extracts. The study also identified active components with cytotoxicity against cancer cells and anti-acetylcholinesterase activity.

Essential Oils of Duguetia Species A. St. Hill (Annonaceae): Chemical Diversity and Pharmacological Potential

Summary: This study reviewed the chemical constituents and pharmacological properties of essential oils (EOs) from Duguetia species. Twelve species were found, and the main chemical constituents reported included bicyclogermacrene and humulene epoxide II. The EOs of Duguetia species exhibited anti-inflammatory, antinociceptive, antibacterial, antifungal, antioxidant, anti-trypanosoma, cytotoxic, and antitumor properties.

Summary: In this study, the cytotoxic and antitumor effects of the essential oil obtained from Guatteria olivacea leaves against liver cancer were evaluated. The essential oil exhibited potential in inhibiting liver cancer growth by inducing apoptosis and DNA fragmentation, without affecting reactive oxygen species levels.

Natural Dibenzo [b,f] oxepines, Pacharin and Bauhiniastatin-1, Isolated from Bauhinia acuruana Induce Apoptosis on Breast Cancer Cells via MCL-1 Protein Reduction

Summary: In this study, the antiproliferative effects of two natural compounds, pacharin and bauhiniastatin-1, isolated from Bauhinia acuruana on breast cancer cells were investigated. The results showed that both compounds exhibited cytotoxicity against various tumor cell lines, including human breast adenocarcinoma, while having no toxicity on normal cells. The mechanism of action was found to be related to the inhibition of myeloid cell leukemia-1 protein, leading to apoptosis of breast adenocarcinoma cells.

Summary: In this study, twenty new acridine-thiosemicarbazone derivatives were synthesized and showed strong antiproliferative activities, with DL-08 exhibiting the highest potency. Certain active derivatives were studied for their interactions with DNA and inhibition of topoisomerase II alpha by in silico and in vitro methods. Non-clinical toxicity tests on mice at a dose of 2000 mg/kg showed no serious signs of toxicity for selected compounds.

Summary: The present study evaluated the therapeutic potential of Arsenic Trioxide (ATO) in treating Oral Squamous Cell Carcinoma (OSCC). The results showed that ATO has antitumoral effects and inhibits the Hedgehog (HH) pathway. Furthermore, ATO also induced changes in cell morphology and cell cycle in oral cancer cells.

Summary: Acute myeloid leukemia (AML) is a heterogeneous disease group with various mutations triggering malignant proliferation. AML relapse is mainly caused by leukemic stem cells (LSCs) with self-renewal capacity and resistance to traditional therapies. LSCs have low oxidative stress levels due to low mitochondrial activity and high ROS-removing pathway activity. Targeting oxidative stress could be a potential strategy to eliminate AML LSCs.

Summary: In this review, the authors provide an overview of the mechanisms of pharmacological action of essential oils and their components in various biological systems. They discuss the antioxidant, anti-inflammatory, antitumor, antimicrobial activities, as well as the mechanisms of action against various types of viruses. The data indicate that the chemical diversity in the composition of essential oils allows them to interfere with biological processes at different levels, making them a promising source for the development of new drugs.

Antimalarials and macrolides: a review of off-label pharmacotherapies during the first wave of the SARS-CoV-2 pandemic

Summary: We critically analyzed clinical trials of chloroquine and hydroxychloroquine with or without macrolides during the first wave of COVID-19 and discussed their limitations. The studies showed that these drugs did not show clinical advantages and were not effective in reducing mortality or improving symptoms in COVID-19 patients. The evidence was based mainly on retrospective trials with low quality, making it unreliable for decision-making.
