Cyclodextrins, since their discovery in the late 19th century, were mainly regarded as excipients. Nevertheless, developments in cyclodextrin research have shown that some of these hosts can capture and include biomolecules, highlighting fatty acids and cholesterol, which implies that they are not inert and that their action may be used in specific medicinal purposes. The present review, centered on literature reports from the year 2000 until the present day, presents a comprehensive description of the known biological activities of cyclodextrins and their implications for medicinal applications. The paper is divided into two main sections, one devoted to the properties and applications of cyclodextrins as active pharmaceutical ingredients in a variety of pathologies, from infectious ailments to cardiovascular dysfunctions and metabolic diseases. The second section is dedicated to the use of cyclodextrins in a range of biomedical technologies.

Inclusion Compound of Efavirenz and γ-Cyclodextrin: Solid State Studies and Effect on Solubility

Susana Santos Braga, Firas El-Saleh, Karyna Lysenko, Filipe A. Almeida Paz

Summary: This study investigated the interaction between gamma-cyclodextrin and efavirenz in both solution and solid state. The 3:2 host-to-guest stoichiometry was found to be the optimal condition for isolating a solid inclusion compound. The effect of gamma-cyclodextrin on the solubility of efavirenz was examined using the isotherm method.

Strategies to Broaden the Applications of Olive Biophenols Oleuropein and Hydroxytyrosol in Food Products

Mariana Monteiro, Andreia F. R. Silva, Daniela Resende, Susana S. Braga, Manuel A. Coimbra, Artur M. S. Silva, Susana M. Cardoso

Summary: Oleuropein and hydroxytyrosol, as health-promoting agents, face challenges in food applications such as degradation and undesired sensory properties. Strategies to overcome these setbacks include encapsulation in delivery systems and covalent and non-covalent complexation. More research is needed to investigate their impact on sensory properties and bioactivity, as well as limitations in the food industry scale.

The Emerging Role of Cyclodextrin Metal-Organic Frameworks in Ostheotherapeutics

Susana Santos Braga, Filipe A. Almeida Paz

Summary: Metal-Organic Frameworks (MOFs), as a class of coordination compounds, have high versatility and loading ability, making them attractive for drug delivery. This review summarizes the early use of cyclodextrin-coated and cyclodextrin-based MOFs in the treatment of bone-related diseases, including cyclodextrin-coated MOFs, cyclodextrin-based MOFs, and cross-linked derivatives of CD-MOFs. The review highlights their applications in delivering bisphosphonates, osteoporotic regulating drugs, and non-steroidal anti-inflammatory drugs for osteoarthritis management, as well as their recent use in dentistry.
