Cancer is a devastating disease that has claimed many lives. Natural bioactive agents from plants are gaining wide attention for their anticancer activities. Several studies have found that natural plant-based bioactive compounds can enhance the efficacy of chemotherapy, and in some cases ameliorate some of the side-effects of drugs used as chemotherapeutic agents. In this paper, we have reviewed the literature on the anticancer effects of four plant-based bioactive compounds namely, curcumin, myricetin, geraniin and tocotrienols (T3) to provide an overview on some of the key findings that are related to this effect. The molecular mechanisms through which the active compounds may exert their anticancer properties in cell and animal-based studies also discussed.

Evaluating Anticancer and Immunomodulatory Effects of Spirulina (Arthrospira) platensis and Gamma-Tocotrienol Supplementation in a Syngeneic Mouse Model of Breast Cancer

Hemavathy Subramaiam, Wan-Loy Chu, Ammu Kutty Radhakrishnan, Srikumar Chakravarthi, Kanga Rani Selvaduray, Yih-Yih Kok

Summary: Nutritional supplements gamma-tocotrienol and Spirulina were evaluated for immune-enhancing and anticancer effects in a mouse model of breast cancer. While individual supplementation with each showed immune modulation effects, the combination did not demonstrate synergistic anticancer effects in this study model.

Tocotrienols protect differentiated SH-SY5Y human neuroblastoma cells against 6-hydroxydopamine-induced cytotoxicity by ameliorating dopamine biosynthesis and dopamine receptor D2 gene expression

Kasthuri Bai Magalingam, Sushela Devi Somanath, Shadab Md, Nagaraja Haleagrahara, Ju-Yen Fu, Kanga Rani Selvaduray, Ammu Kutty Radhakrishnan

Summary: This study demonstrates the neuroprotective effects of T3 and its analogues against oxidative damage in Parkinson's disease, with TRF showing significant protection of dopamine neurons. Gamma-T3 was most effective in attenuating apoptosis, while alpha-T3 showed the most efficient antioxidant effects.

Clinically Relevant Genes and Proteins Modulated by Tocotrienols in Human Colon Cancer Cell Lines: Systematic Scoping Review

Ali Qusay Khalid, Saatheeyavaane Bhuvanendran, Kasthuri Bai Magalingam, Premdass Ramdas, Mangala Kumari, Ammu Kutty Radhakrishnan

Summary: The research in the past decade has shown that tocotrienols have potent anti-cancer properties, particularly in the field of oncology, and may have important therapeutic effects on various cancers such as colorectal cancer. Bioactive natural compounds like tocotrienols show promise in preventing tumor and cancer progression. Studies on tocotrienols and human CRC cells have focused on exploring their modulation of gene expression and encoded proteins, as well as their mechanisms in intervening cancer progression.

6-Hydroxydopamine Induces Neurodegeneration in Terminally Differentiated SH-SY5Y Neuroblastoma Cells via Enrichment of the Nucleosomal Degradation Pathway: a Global Proteomics Approach

Kasthuri Bai Magalingam, Sushela Devi Somanath, Premdass Ramdas, Nagaraja Haleagrahara, Ammu Kutty Radhakrishnan

Summary: This study investigated the protein expression changes in SH-SY5Y cells induced by 6-OHDA and their role in Parkinson's disease. The results suggest that abnormal neuronal development and ribosome pathway may play important roles in the neurodegeneration of PD, while heat shock proteins and histone release are also involved in the pathogenesis of PD.

Clinical characteristics, cytokine profiles and plasma IgE in adults with asthma

Rathimalar Ayakannu, Nor Azizan Abdullah, Vijaya Lechimi Raj, Ammu K. Radhakrishnan, Chong Kin Liam

Summary: This study explored the clinical characteristics, cytokine profile, and plasma IgE levels in Malaysian asthma patients. The results showed that asthmatics had higher levels of plasma IgE and cytokines compared to controls. However, there was no significant association between cytokine levels and asthma severity or clinical symptoms.

Dina El-Rabie Osman, Brandon Wee Siang Phon, Muhamad Noor Alfarizal Kamarudin, Stephen Navendran Ponnampalam, Ammu Kutty Radhakrishnan, Saatheeyavaane Bhuvanendran

Summary: This paper systematically reviews and describes the roles of various biomarkers regulated by lipid-soluble vitamins in the pathophysiology of GBM. The results suggest that retinoids activate cell differentiation, vitamin D modulates cell cycle biomarkers, and vitamin E derivatives modulate apoptosis-related biomarkers.

Tocotrienol-Rich Fraction and Levodopa Regulate Proteins Involved in Parkinson's Disease-Associated Pathways in Differentiated Neuroblastoma Cells: Insights from Quantitative Proteomic Analysis

Kasthuri Bai Magalingam, Premdass Ramdas, Sushela Devi Somanath, Kanga Rani Selvaduray, Saatheeyavaane Bhuvanendran, Ammu Kutty Radhakrishnan

Summary: This study investigated the proteomic changes in human neural cells induced by TRF, a palm oil-derived vitamin E fraction, using a label-free LC-MS/MS approach. The results showed that TRF and levodopa, a drug used in Parkinson's disease treatment, regulated the expression of a number of proteins involved in crucial pathways such as ubiquitin-proteasome, calcium signaling, protein processing in the endoplasmic reticulum, mitochondrial pathway, and axonal transport system. Overall, TRF is an important functional food that affects protein expression in human neuronal cells.

Identification of Prominent Genes between 3D Glioblastoma Models and Clinical Samples via GEO/TCGA/CGGA Data Analysis

Brandon Wee Siang Phon, Saatheeyavaane Bhuvanendran, Qasim Ayub, Ammu Kutty Radhakrishnan, Muhamad Noor Alfarizal Kamarudin

Summary: This study investigated the suitability of 3D cultures as cell-based models for GBM by leveraging genomic data. Genes related to pathways such as epithelial-to-mesenchymal transition, angiogenesis/migration, hypoxia, stemness, and the Wnt signalling pathway were found to be upregulated in both brain samples from GBM patients and 3D GBM cells. These findings support the use of 3D GBM cultures as reliable preclinical models for studying GBM.

Protease-targeting peptide-functionalized porous silicon nanoparticles for cancer fluorescence imaging

Jayasree S. Kanathasan, Uma Devi Palanisamy, Ammu K. Radhakrishnan, Srikumar Chakravarthi, Tan Boon Thong, Varghese Swamy

Summary: This study investigates the targeting potential of pSi NPs functionalized with two types of peptides and finds that the linear peptide-pSi NP conjugate has better stability and higher binding efficiency under tumor conditions. In vivo studies show a higher accumulation of linear peptide-conjugated pSi NPs in the tumor site. These results suggest that the linear peptide-pSi NP formulation is a potential drug-delivery platform and fluorescence bioimaging agent.
