Natural Sulfur-Containing Compounds: An Alternative Therapeutic Strategy against Liver Fibrosis

Liver fibrosis is a pathophysiologic process involving the accumulation of extracellular matrix proteins as collagen deposition. Advanced liver fibrosis can evolve in cirrhosis, portal hypertension and often requires liver transplantation. At the cellular level, hepatic fibrosis involves the activation of hepatic stellate cells and their transdifferentiation into myofibroblasts. Numerous pro-fibrogenic mediators including the transforming growth factor-beta 1, the platelet-derived growth factor, endothelin-1, toll-like receptor 4, and reactive oxygen species are key players in this process. Knowledge of the cellular and molecular mechanisms underlying hepatic fibrosis development need to be extended to find novel therapeutic strategies. Antifibrotic therapies aim to inhibit the accumulation of fibrogenic cells and/or prevent the deposition of extracellular matrix proteins. Natural products from terrestrial and marine sources, including sulfur-containing compounds, exhibit promising activities for the treatment of fibrotic pathology. Although many therapeutic interventions are effective in experimental models of liver fibrosis, their efficacy and safety in humans are largely unknown. This review aims to provide a reference collection on experimentally tested natural anti-fibrotic compounds, with particular attention on sulfur-containing molecules. Their chemical structure, sources, mode of action, molecular targets, and pharmacological activity in the treatment of liver disease will be discussed.

Metabolic Adaptations to Marine Environments: Molecular Diversity and Evolution of Ovothiol Biosynthesis in Bacteria

Summary: Research identified the distribution of ovothiol biosynthetic enzymes in bacteria through genomic and metagenomic data mining. These enzymes are mainly found in marine aerobic Proteobacteria, some of which establish relationships with other organisms. A horizontal gene transfer event of ovothiol biosynthetic enzymes was also discovered.

Researchers are exploring the potential of AMPs and their derivatives as therapies against COVID-19, considering their interactions with toll-like receptors (TLRs) and chemokine receptors in the context of the disease's pathogenesis. Additionally, controlled physical activity, along with a healthy lifestyle, may serve as natural defense against COVID-19 and support therapies, as seen in the increase of serum defensins in competitive athletes.

Challenges and advances towards the rational design of microalgal synthetic promoters in Chlamydomonas reinhardtii

Summary: This review focuses on Chlamydomonas reinhardtii, a microalgal species commonly used in bioengineering research, and discusses the key obstacles and potential solutions in synthetic promoter development. Microalgae have great potential as carbon-fixing biofactories to provide safe and sustainable high-value compounds, but optimizing their metabolic outputs and making them competitive with established industrial hosts requires precise control over transgene expression. However, the current repertoire of functional synthetic promoters for microalgae, especially for C. reinhardtii, is limited compared to other commercial chassis. The review highlights the need to expand the microalgal gene expression toolbox and proposes novel approaches and perspectives for synthetic promoter development in microalgae.
