Conservation and Divergence in the Meiocyte sRNAomes of Arabidopsis, Soybean, and Cucumber

Meiosis is a critical process for sexual reproduction. During meiosis, genetic information on homologous chromosomes is shuffled through meiotic recombination to produce gametes with novel allelic combinations. Meiosis and recombination are orchestrated by several mechanisms including regulation by small RNAs (sRNAs). Our previous work in Arabidopsis (Arabidopsis thaliana) meiocytes showed that meiocyte-specific sRNAs (ms-sRNAs) have distinct characteristics, including positive association with the coding region of genes that are transcriptionally upregulated during meiosis. Here, we characterized the ms-sRNAs in two important crops, soybean (Glycine max) and cucumber (Cucumis sativus). Ms-sRNAs in soybean have the same features as those in Arabidopsis, suggesting that they may play a conserved role in eudicots. We also investigated the profiles of microRNAs (miRNAs) and phased secondary small interfering RNAs in the meiocytes of all three species. Two conserved miRNAs, miR390 and miR167, are highly abundant in the meiocytes of all three species. In addition, we identified three novel cucumber miRNAs. Intriguingly, our data show that the previously identified phased secondary small interfering RNA pathway involving soybean-specific miR4392 is more abundant in meiocytes. These results showcase the conservation and divergence of ms-sRNAs in flowering plants, and broaden our understanding of sRNA function in crop species.

Spatiotemporal formation of the large vacuole regulated by the BIN2-VLG module is required for female gametophyte development in Arabidopsis

Summary: In Arabidopsis thaliana, the development of female gametophyte (FG) involves the formation and expansion of large vacuoles. The VACUOLELESS GAMETOPHYTES (VLG) gene is responsible for facilitating the fusion of vesicles to form these vacuoles, but its regulation is still not well understood. This study found that mutations in the BRASSINOSTEROID INSENSITIVE2 (BIN2) gene increased the abundance of VLG, leading to excessive vacuole formation and FG abortion. On the other hand, mutations in BIN2 and its homologs reduced VLG abundance and caused phenotypes similar to vlg/VLG mutants. It was also discovered that BIN2 interacts with and phosphorylates VLG, enhancing its stability and abundance to promote vacuole formation.

Harnessing Wolbachia cytoplasmic incompatibility alleles for confined gene drive: A modeling study

Summary: Wolbachia are maternally-inherited bacteria that can rapidly spread in populations by manipulating reproduction. The cifA and cifB genes found in Wolbachia phage play a crucial role in cytoplasmic incompatibility, the most common form of Wolbachia reproductive interference. By utilizing these genes, new toxin-antidote gene drives can be constructed in insect genomes, which can limit or confine the spread of desired traits in populations. These drives are safe and flexible for population modification of insects and offer a powerful tool for genetic modification.

Ubiquitination of stalled ribosomes enables mRNA decay via HBS-1 and NONU-1 in vivo

Summary: Ribosomes are responsible for reading mRNAs and synthesizing proteins. When ribosomes encounter traffic jams on mRNAs, cells have mechanisms to clean up the jam and remove problematic mRNAs. In this study, the machinery for handling ribosome stalls and the relationships between factors were identified in C. elegans. It was found that ribosomes need to be tagged by ubiquitin for mRNA decay, and the absence of ubiquitin sites on ribosomes prevents mRNA decay. Additionally, an enzyme that cuts mRNA is recruited by ubiquitin signals, and ribosome rescue factors are also required for mRNA decay.

Visualizing cell-cell communication using synthetic notch activated MRI

Summary: Cell-cell communication is crucial in multicellular organisms, especially in cell-based cancer immunotherapies where engineered immune cells interact with cancer cells through specific antigens. By using the synthetic Notch system, T cells engineered in this study were able to induce the expression of reporter genes in response to interaction with a chosen antigen on neighboring cancer cells. The high spatial resolution and tomographic nature of MRI allowed for clear visualization and mapping of the engineered cells within tumors, showing promise for monitoring cell therapies in patients and understanding interactions between cell populations in the body.

Risk factors and new diagnostic index for deep venous thrombosis of lower extremities in elderly patients with traumatic femoral neck fracture

Summary: This study aimed to determine the incidence and risk factors of deep vein thrombosis (DVT) in elderly Chinese patients with femoral neck fracture at admission, and to establish and evaluate a new DVT predictive index based on these risk factors. The results showed that the injured side, hemoglobin, fibrinogen, and d-dimer were independent risk factors for DVT occurrence. The new predictive index based on these factors had a high diagnostic value.

Nuclear phylogeny and insights into whole- genome duplications and reproductive development of Solanaceae plants

Detection of Doxycycline Using Carbon Quantum dots as Probe Based on Internal Filtering Effect
