Line Scan Raman Microspectroscopy for Label-Free Diagnosis of Human Pituitary Biopsies

Pituitary adenomas are neoplasia of the anterior pituitary gland and can be subdivided into hormone-producing tumors (lactotroph, corticotroph, gonadotroph, somatotroph, thyreotroph or plurihormonal) and hormone-inactive tumors (silent or null cell adenomas) based on their hormonal status. We therefore developed a line scan Raman microspectroscopy (LSRM) system to detect, discriminate and hyperspectrally visualize pituitary gland from pituitary adenomas based on molecular differences. By applying principal component analysis followed by a k-nearest neighbor algorithm, specific hormone states were identified and a clear discrimination between pituitary gland and various adenoma subtypes was achieved. The classifier yielded an accuracy of 95% for gland tissue and 84-99% for adenoma subtypes. With an overall accuracy of 92%, our LSRM system has proven its potential to differentiate pituitary gland from pituitary adenomas. LSRM images based on the presence of specific Raman bands were created, and such images provided additional insight into the spatial distribution of particular molecular compounds. Pathological states could be molecularly differentiated and characterized with texture analysis evaluating Grey Level Cooccurrence Matrices for each LSRM image, as well as correlation coefficients between LSRM images.

Quantification of intrinsic optical signals in the outer human retina using optical coherence tomography

Summary: In this study, the use of intrinsic optical signals as a noninvasive biomarker for assessing retinal photoreceptor function was explored. Results showed dynamic changes in optical path length between different anatomical structures in the outer retina during light stimulation, suggesting the potential for using a standard OCT platform and signal model in measuring subretinal changes in response to light adaptation.

Elevated EMMPRIN Serum Levels Indicate Plaque Vulnerability in Patients With Asymptomatic High Grade Carotid Stenosis

Summary: This study aimed to test extracellular matrix metalloproteinase inducer (EMMPRIN) as a potential marker of high risk vulnerable plaques in patients with carotid artery stenosis. EMMPRIN levels were analyzed in pre-operative serum samples and it was found that patients with high risk plaques had increased levels of EMMPRIN. The combination of EMMPRIN with clinical risk factors improved the prediction of vulnerable plaques in asymptomatic patients.

Does High-Definition 3-Dimensional Imaging Improve Orientation During Endoscopic Transsphenoidal Surgery? A Prospective Trial

Ultra-high-resolution optical coherence tomography for the investigation of thin multilayered pharmaceutical coatings

Summary: Optical Coherence Tomography (OCT) has gained attention as a promising technology for monitoring pharmaceutical film-coating processes and end-point detection. We present an ultra-high-resolution (UHR-) OCT and demonstrate its performance on different multiparticulate dosage forms with different layer structures. The achieved system resolution enables the assessment of defects, film thickness variability, and morphological features within the coating.

Validation of Myc-Associated Protein X (MAX) regulation in growth hormone secreting and nonfunctional pituitary adenoma

Summary: This study aimed to validate the differential DNA methylation and MAX protein expression profiles between nonfunctional pituitary adenoma (NFPA) and growth hormone-secreting pituitary adenoma (GHPA). Results showed that GHPA exhibited more hypomethylation events across all known MAX binding sites, with 1,551 sites having significantly different methylation patterns near potentially MAX-regulated promoter regions. Gene ontology analysis suggested involvement in cellular proliferation, tumor invasion, and hormonal secretion. GHPA also demonstrated significantly increased expression of MAX protein compared to NFPA.

Ectopic lipid metabolism in anterior pituitary dysfunction

Summary: In industrialized countries, changes in lifestyle and diet have caused an increase in obesity and metabolic disorders. Excessive lipid deposition in organs and tissues with limited capacity for lipid storage disrupts metabolic function and increases the risk of metabolic diseases and complications. Pituitary disorders can indirectly affect ectopic lipid deposition by altering lipid metabolism and insulin sensitivity, as well as directly influencing energy metabolism through specific hormonal effects. This review provides information on the impact of pituitary disorders on ectopic fat stores and explores the potential pathophysiological mechanisms involved in hormone action in ectopic lipid metabolism.
