Intrinsic connectomes are a predictive biomarker of remission in major depressive disorder

Although major depressive disorder (MDD) is associated with altered functional coupling between disparate neural networks, the degree to which such measures are ameliorated by antidepressant treatment is unclear. It is also unclear whether functional connectivity can be used as a predictive biomarker of treatment response. Here, we used whole-brain functional connectivity analysis to identify neural signatures of remission following antidepressant treatment, and to identify connectomic predictors of treatment response. 163 MDD and 62 healthy individuals underwent functional MRI during pre-treatment baseline and 8-week follow-up sessions. Patients were randomized to escitalopram, sertraline or venlafaxine-XR antidepressants and assessed at follow-up for remission. Baseline measures of intrinsic functional connectivity between each pair of 333 regions were analyzed to identify pre-treatment connectomic features that distinguish remitters from non-remitters. We then interrogated these connectomic differences to determine if they changed post-treatment, distinguished patients from controls, and were modulated by medication type. Irrespective of medication type, remitters were distinguished from non-remitters by greater connectivity within the default mode network (DMN); specifically, between the DMN, fronto-parietal and somatomotor networks, the DMN and visual, limbic, auditory and ventral attention networks, and between the fronto-parietal and somatomotor networks with cingulo-opercular and dorsal attention networks. This baseline hypo-connectivity for non-remitters also distinguished them from controls and increased following treatment. In contrast, connectivity for remitters was higher than controls at baseline and also following remission, suggesting a trait-like connectomic characteristic. Increased functional connectivity within and between large-scale intrinsic brain networks may characterize acute recovery with antidepressants in depression.

Summary: A genome-wide association study identified significant genetic loci associated with suicide attempts, particularly one locus on chromosome 7 which is implicated in risk-taking behavior, smoking, and insomnia. The study also revealed strong genetic correlations between suicide attempts and psychiatric disorders, as well as other nonpsychiatric traits like smoking, pain, and lower socioeconomic status. Conditioning on psychiatric disorders reduced the genetic correlations between suicide attempts and psychiatric disorders, indicating a shared biology with nonpsychiatric risk factors.

Moral Injury, Traumatic Stress, and Threats to Core Human Needs in Health-Care Workers: The COVID-19 Pandemic as a Dehumanizing Experience

Summary: The pandemic has posed a threat to core human needs, especially in terms of meeting basic human needs and moral incongruence resulting in psychological injury. This study found that nearly one third of surveyed healthcare workers reported symptoms of psychological trauma, and the prevalence of suicidal ideation was roughly three times higher among healthcare workers compared to the general population. Moral injury and loneliness were found to predict greater symptoms of traumatic stress and suicidality. The pandemic primarily threatened basic human motivations at the foundational level of safety and security relative to other higher order needs.

Attention enhances category representations across the brain with strengthened residual correlations to ventral temporal cortex

Summary: This study used fMRI to investigate how attention modulates neural representations of goal-relevant stimuli in the brain. The results showed that when two objects are simultaneously viewed, the category of the attended object can be more readily decoded. After accounting for stimulus-driven variance, the correlation in residual brain activity between a cortical region and a category-selective region of VTC was higher when the preferred category was attended. This correlation was particularly strong in the right occipital, parietal, and frontal cortices. Furthermore, stronger residual correlations between a given region and VTC were associated with better visual category information decoding.

Grey matter covariation and the role of emotion reappraisal in mental wellbeing and resilience after early life stress exposure

Summary: Resilience is a crucial process of adaptive recovery for maintaining mental wellbeing after stress exposure. This study investigated the association between resilience and grey matter covariation (GMC) in healthy adults, with and without early life stress (ELS) exposure, and examined the mediating role of emotion regulation. The findings showed distinct morphometric patterns related to resilience in individuals with ELS exposure. Decreased GMC in the temporo-parietal regions was associated with wellbeing in participants without ELS exposure, while increased patterns of covariation in regions related to salience and executive control networks, as well as decreased GMC in temporo-parietal areas, were associated with resilience in ELS-exposed participants. Cognitive reappraisal mediated the brain-wellbeing relationship only in ELS-exposed participants. The study suggests that stronger GMC in regions associated with emotion and cognitive functioning in ELS-exposed participants with high wellbeing may indicate neural signatures of resilience, potentially enhanced by adaptive emotion regulation.

Cross-trial prediction of depression remission using problem-solving therapy: A machine learning approach

Valence processing alterations in SAPAP3 knockout mice and human OCD
