A botanical drug composed of three herbal materials attenuates the sensorimotor gating deficit and cognitive impairment induced by MK-801 in mice

Objectives A botanical drug derived from the ethanolic extract composed of Clematis chinensis Osbeck (Ranunculaceae), Trichosanthes kirilowii Maximowicz (Cucurbitaceae) and Prunella vulgaris Linne (Lamiaceae) has been used to ameliorate rheumatoid arthritis as an ethical drug in Korea. In our study, we investigated the effect of this herbal complex extract (HCE) on schizophrenia-like behaviours induced by MK-801. Methods HCE (30, 100 or 300 mg/kg, p.o) was orally administered to male ICR mice to a schizophrenia-like animal model induced by MK-801. We conducted an acoustic startle response task, an open-field task, a novel object recognition task and a social novelty preference task. Key findings We found that a single administration of HCE (100 or 300 mg/kg) ameliorated MK-801-induced abnormal behaviours including sensorimotor gating deficits and social or object recognition memory deficits. In addition, MK-801-induced increases in phosphorylated Akt and GSK-3 beta expression levels in the prefrontal cortex were reversed by HCE (30, 100 or 300 mg/kg). Conclusions These results imply that HCE ameliorates MK-801-induced dysfunctions in prepulse inhibition, social interactions and cognitive function, partly by regulating the Akt and GSK-3 beta signalling pathways.

A Deep Dive: SIWV Tetra-Peptide Enhancing the Penetration of Nanotherapeutics into the Glioblastoma

Summary: GBM is an aggressive malignant tumor that is difficult to regulate with conventional chemotherapy due to anatomical structure specificity, low drug targeting ability, and limited penetration depth. A study prepared four types of nanoparticles based on porous silicon nanoparticles incorporating PEG, iRGD peptide, and SIWV tetra-peptide, and evaluated their deep-tumor penetration abilities in different models. The SIWV tetra-peptide significantly enhanced the penetration depth of the nanoparticles and showed higher anticancer efficacy in therapeutic formulations, promising for nanotherapeutics and peptide-conjugated drugs for GBM.

4-(2-Hydroxyethyl)-1-piperazine ethane sulfonic acid repositioning: Amyloid disaggregating agent and its sustained-release system

Summary: In this study, we found that HEPES is capable of disaggregating amyloid plaques associated with Alzheimer's disease. Additionally, we demonstrated its sustained-release system for up to 2 weeks through hybridization with a biocompatible polymer, Eudragit S100.

Effects of repetitive training on learning and memory performance of TLR2 KO mice

Summary: Current antipsychotics have limited effects on the cognitive deficits of schizophrenia patients. In this study, a Toll-like receptor knockout mouse was used as a schizophrenia mouse model to evaluate the effects of repetitive training as a cognitive remediation therapy. The results showed that repetitive training improved memory retrieval and reversal learning ability in the Toll-like receptor knockout mice, and activated neuronal cells in specific brain regions. This study suggests that the Toll-like receptor knockout mouse is a useful tool for studying the neurobiological mechanisms of cognitive remediation in schizophrenia.

First-in-Class: Cervical cancer diagnosis based on a urine test with fluorescent cysteine probe

Summary: In this study, the world's first in vitro cervical cancer diagnosis technology based on a urine test was developed and its effectiveness was verified. The fluorescence-based diagnostic method showed a high true-positive ratio (74%) for cervical cancer. It has advantages of simple equipment, small sample requirement, and short diagnosis time.

TAMRA-conjugated DRL tripeptide for the visualization of synovium

Summary: A new synovium-homing tri-peptide, DRL, was developed through peptide screening and functionalized to visualize the synovium. The TAMRA-conjugated DRL, named DRL-TAM, demonstrated remarkable visualization ability on synovial-derived cells and synovial-related organs in cell screening and in vivo fluorescence imaging studies. This study opens up new possibilities for the diagnosis and treatment of synovium-related diseases.

All-Nontoxic Fluorescent Probe for Biothiols and Its Clinical Applications for Real-Time Glioblastoma Visualization

Summary: Fluorescence-guided surgery (FSG) is a surgical method that uses fluorescent materials to selectively visualize tumor sites. However, commercially available fluorescent materials have shown serious adverse effects. A new nontoxic fluorescent probe called NPO-o-Pyr has been developed for real-time human glioblastoma visualization. It has high specificity and sensitivity, fast response time, and high biocompatibility. NPO-o-Pyr can overcome the limitations of current fluorescent probes and become a new substitute in FSG.

Potential Alzheimer's disease therapeutic nano-platform: Discovery of amyloid-beta plaque disaggregating agent and brain-targeted delivery system using porous silicon nanoparticles

Summary: Recent research has shown that using porous silicon nanoparticles as a therapeutic platform can overcome Alzheimer's disease and deliver drugs to the brain. The study found that 6-amino-2-naphthalenesulfonic acid (ANA) is an effective compound that can disaggregate amyloid plaques. By loading ANA into porous silicon nanoparticles and targeting delivery, memory impairment in an Alzheimer's disease mouse model can be improved.
