Micro solid-phase extraction for the analysis of per- and polyfluoroalkyl substances in environmental waters

Growing concern over the environmental and health impacts of per- and polyfluoroalkyl substances (PFASs) has led to the development of increasingly stringent regulatory guidelines. To meet these guidelines for the determination of PFASs in surface-water, solid-phase extraction (SPE) is commonly implemented for clean-up and pre-concentration of samples. In this paper a micro-SPE method for the cleanup and pre-concentration of PFASs from surface-water was developed. A micro-SPE packing phase was created to retain 13 long and short chain PFAS after examining combinations of four 3 mu m particle size sorbents, with the optimal phase consisting of a 50:50 mixture of C18 and aminopropyl silica. Micro-SPE achieved similar results to conventional SPE methods while reducing sample preparation time to 5 min and using only 2 mL of sample. The method was validated using spiked recoveries (100 ng L-1) from PFAS contaminated surface-water samples with recoveries ranging from 86% to 111% and relative standard deviations below 18%. Concentrations of the PFASs in the samples ranged from below the limit of quantification to 898 +/- 15 ng L-1. Automation of sample preparation, including the micro-SPE extraction, was also demonstrated. These results show the potential for automated micro-SPE to replace conventional SPE, with the decreases in sample preparation time, sample and solvent volumes crucial for incorporation into routine analyses in commercial laboratories. Preparation of matrix-matched standards for the analysis of teeth via laser ablation-inductively coupled plasma-mass spectrometry

Towards Non-Targeted Screening of Lipid Biomarkers for Improved Equine Anti-Doping

Optimization of chromatographic buffer conditions for the simultaneous analysis of phosphatidylinositol and phosphatidylinositol phosphate species in canola

Summary: Phosphatidylinositols and phosphatidylinositol phosphates are closely related lipids that have been found to impact various cellular functions and are correlated with the development and progression of diseases. The analysis of these compounds is challenging due to their unique chemical characteristics, and current lipidomics methods are unsuitable. In this study, a new method was developed to enable the sensitive and simultaneous analysis of phosphatidylinositols and phosphatidylinositol phosphates, allowing for the identification of 148 species. This analysis was able to differentiate four distinct canola cultivars based on their phosphatidylinositide-lipidome, suggesting the potential use of lipidomic profiles in studying disease development and progression.

The toxic metal hypothesis for neurological disorders

Summary: Multiple sclerosis and major neurodegenerative disorders may have both genetic and environmental causes, with toxic metals being implicated as potential triggers for these conditions. This hypothesis suggests that damage to selective neurons from toxic metals causes dysfunction of the blood-brain barrier, leading to the entry of circulating toxicants that damage cells in the nervous system. The type of neurological disorder that arises depends on various factors, including which neurons are damaged, genetic variants, exposure levels and durations, and the mixture of toxic metals. Further research is needed to support this hypothesis, but taking steps to reduce environmental toxic metal pollution is recommended to protect the nervous system.

Immunolabelling perturbs the endogenous and antibody-conjugated elemental concentrations during immuno-mass spectrometry imaging

Summary: Immuno-mass spectrometry imaging is a method used to quantitatively analyze the spatial distribution of biomolecules using laser ablation-inductively coupled plasma-mass spectrometry (LA-ICP-MS) and lanthanide-conjugated antibodies. The effects of immunolabelling and coverslipping on the concentrations of endogenous metals and Gd-conjugated antibodies were investigated using cryofixed muscle sections. Immunolabelling steps were found to impact the concentration of endogenous elements, and separate histological sections were recommended for quantitative analysis. Additionally, organic mounting media were suggested for coverslipping tissue sections to maintain the integrity of the elemental label.

Concentration and Distribution of Toxic and Essential Elements in Traditional Rice Varieties of Sri Lanka Grown on an Anuradhapura District Farm

Summary: Toxic heavy metals, including arsenic, cadmium, and lead, may play a role in the development of chronic kidney disease of unknown etiology (CKDu) in Sri Lanka. This study investigated the concentrations of essential and toxic elements in traditional rice varieties regularly grown in a CKDu hotspot. The results showed potentially unsafe concentrations of cadmium in one store-bought sample and revealed the distribution of lead and cadmium in the rice grains. Traditional rice varieties were found to have higher levels of essential elements compared to non-traditional varieties.

Summary: This study presents a newly developed and validated method for the detection and quantification of BMAA, its isomers, and major MCs using RP LC-MS/MS. The application of this method on a culture of Microcystis aeruginosa resulted in the simultaneous detection of 2,4-DAB and MC-YR. The study provides a unified method for the quantitative analysis of these cyanotoxins in natural environmental samples.

Effects of the Toxic Non-Protein Amino Acid β-Methylamino-L-Alanine (BMAA) on Intracellular Amino Acid Levels in Neuroblastoma Cells

Summary: The cyanobacterial neurotoxin BMAA had a profound effect on intracellular amino acid levels in human neuroblastoma cells at specific time points, but homeostasis was restored in most cases. Supplementation with L-serine protected against BMAA toxicity. Amino acids potentially involved in protection against BMAA-induced oxidation were depleted in cells at later time points.

Spatial distribution of trace metals and associated transport proteins during bacterial infection

Breakthrough curve analysis of phosphorylated hazelnut shell waste in column operation for continuous harvesting of lithium from water

Summary: Biosorption using phosphorylated hazelnut shell waste (FHS) is a promising method for lithium removal and recovery. This study investigated the performance of a continuous-flow packed-bed column system using FHS for ultra-low level lithium removal. The results showed that the lihium biosorption capacity of the FHS column was unaffected by the bed height, but decreased with increasing flow rate.

Multivariate surface self-assembly strategy to fabricate ionic covalent organic framework surface grafting monolithic sorbent for enrichment of aristolochic acids prior to high performance liquid chromatography analysis

Summary: In this study, an ionic covalent organic framework (iCOF) surface grafting monolithic sorbent was prepared for in-tube solid-phase microextraction (SPME) of aristolochic acids (AAs) in various samples. The density of quaternary ammonium ions in the iCOF was modulated for enhanced adsorption of AAs. The iCOF surface grafting monolithic sorbent was confirmed by different means. Several operating conditions were optimized to develop the online in-tube SPME-HPLC method, which showed good linearity, low limits of detection, and high recoveries for AAs in serum samples. This study presents a feasible approach for the application of COF-based monolithic sorbents in sample preparation.

Chitosan-alginate sponge with multiple cross-linking networks for adsorption of anionic dyes: Preparation, property evaluation, mechanism exploration, and application

Summary: A chitosan-alginate sponge with multiple cross-linking networks was developed for adsorption and enrichment of anionic dyes from food samples. The sponge showed better three-dimensional network structure, mechanical behavior, and stability compared to pure chitosan and alginate sponges. The adsorption performance and selectivity of the sponge were systematically studied, and it maintained high adsorption capacity and selectivity for anionic dyes. The sponge also showed satisfactory reusability and was successfully combined with high-performance liquid chromatography for the enrichment and detection of anionic dyes in candy and cocktail samples.

An integrated approach for studying the metabolic profiling of herbal medicine in mice using high-resolution mass spectrometry and metabolomics data processing tools

Summary: Analysis of in vivo exposure to traditional Chinese medicine using mass spectrometry is helpful for identifying effective ingredients and developing new drugs. This study focused on Forsythia suspensa (Lian Qiao) and used metabolomics technology to explore its in vivo exposure. By comparing the differences before and after administration, a complete set of nontargeted analysis strategies for TCM exposure was established. The study identified 393 Lian Qiao-related chemical components in mice after administration, including prototypes and metabolites. This study provides a reference for research on the active ingredients of Lian Qiao in vivo.

A two-run heart-cut multidimensional gas chromatography method using flame ionization and mass spectrometry for automated and robust determination of nearly complete wine aroma-volatile profiles

Summary: A quantitative analytical method was developed and validated in this study to determine the concentrations of 81 aroma-relevant wine volatiles. The method utilizes stir bar sorptive extraction and multidimensional gas chromatography with flame ionization detector or mass spectrometer. The method demonstrated good linearity, reproducibility, and recovery, and was successfully applied to analyze 20 wine samples.
