A range of novel cyclometalated ruthenium(II) and iridium(III) complexes with a steroidal backbone based on androsterone were synthesized and characterized by NMR spectroscopy and X-ray crystallography. Their cytotoxic properties in RT112 and RT112 cP (cisplatin-resistant) cell lines as well as in MCF7 and somatic fibroblasts were compared with those of the corresponding nonsteroidal complexes and the noncyclometalated pyridyl complexes as well as with cisplatin as reference. All steroidal complexes were more active in RT112 cP cells than cisplatin, whereby the cyclometalated pyridinylphenyl complexes based on 5c showed high cytotoxicity while maintaining low resistant factors of 0.33 and 0.50.

Controlled, Stepwise Syntheses of Oligomers with Modified Quinoxaline Backbones

Jerome Klein, Nicole Jung, Stefan Braese

Summary: New oligomers based on quinoxaline units were synthesized through multistep reactions, using Wittig coupling to yield (E)-(quinoxalin-2-yl)ethene oligomers. Diverse quinoxaline-based phosphonium salts were designed and synthesized, providing versatility and compatibility in the oligomer-building process. Characterization of the oligomers revealed excellent stereoisomer specificity, with a fully E-configurated conjugated pi-system. The light absorption/emission profiles of the oligomers indicate potential applications in materials science.

Practical synthesis and biological screening of sulfonyl hydrazides

Joao Macara, Catarina Caldeira, Jose Cunha, Jaime A. S. Coelho, Maria J. S. A. Silva, Konrad Kramer, Christoph W. Grathwol, Stefan Brase, M. Manuel B. Marques

Summary: A methodology for the synthesis of sulfonyl hydrazides mediated by hypervalent iodine is described. The polarity of sodium sulfinate salts is reversed by hypervalent iodine reagents to generate a key intermediate, which can react with mono- and disubstituted hydrazines. Various sulfonyl hydrazides were successfully synthesized with yields up to 62%, demonstrating the practical utility of this protocol. Mechanistic studies, including DFT calculations, were performed and the bioactivity of the synthesized compounds was evaluated.

Challenges and Opportunities in 3D Laser Printing Based on (1+1)-Photon Absorption

Vincent Hahn, N. Maximilian Bojanowski, Pascal Rietz, Florian Feist, Mariana Kozlowska, Wolfgang Wenzel, Eva Blasco, Stefan Braese, Christopher Barner-Kowollik, Martin Wegener

Summary: Most light-based 3D printing methods rely on optical or chemical nonlinearities for polymerization reactions. Two-photon absorption is commonly used in 3D micro-and nanoprinting, but this article reviews three emerging processes that allow for quadratic nonlinearity without relying on two-photon absorption, namely upconversion luminescence, two-step absorption, and triplet-triplet annihilation. These (1 + 1)-photon absorption processes enable the use of continuous-wave lasers at lower peak powers and cost compared to two-photon absorption. The article discusses recent progress, challenges, and future perspectives.

Membrane-acting biomimetic peptoids against visceral leishmaniasis

Vivek Kumar, Jennifer S. Lin, Natalia Molchanova, John A. Fortkort, Carolin Reckmann, Stefan Braese, Havard Jenssen, Annelise E. Barron, Archana Chugh

Summary: Visceral leishmaniasis (VL) is a neglected tropical disease and the use of cell-permeating peptides as potential drug candidates to treat VL is limited due to their limited biological stability. In this study, peptoids, which are peptide mimics with good biostability, were tested against Leishmania donovani. The addition of N-alkyl hydrophobic chains and bromination yielded compounds with good antileishmanial activity against both intracellular amastigotes and promastigotes, indicating the potential of these peptoids as drug candidates for VL.

Stereoselective semisynthesis of uzarigenin and allo-uzarigenin

Sarah Al Muthafer, Christoph Schissler, Vanessa Koch, Hannes Kuhner, Martin Nieger, Stefan Brase

Summary: In this study, the concise semisynthesis of natural cardenolide uzarigenin and its diastereoisomer allo-uzarigenin was achieved in nine and seven steps, respectively, starting from epi-androsterone. The stereoselective introduction of the beta-hydroxy group at C-14 via Mukaiyama oxidation and the installation of the butenolide ring at C-17 using a Stille-cross-coupling reaction were discussed. By directing the hydrogenation via a protecting group strategy, both the C-17 beta isomer and allo-uzarigenin were obtained stereoselectively.

Summary: This study presents a method to screen for selective peptide binders to perfluorinated sulfonic acid ionomers, such as Nafion, using ultra-high density peptide arrays. The peptide WIWHCW with a helix structure is identified as a selective binder to Nafion, and the invariant amino acids responsible for binding are also determined. The dissociation constant of WIWHCW to Nafion is measured at approximately 140 μM.

Valentina Ferraro, Clara R. Adam, Aleksandra Vranic, Stefan Braese

Summary: The use of visible light for polymerization is appealing for its potential in improving the sustainability of the process. Transition metal complexes, with their long-lived excited states and reversible redox properties, are a viable option for photoinitiating systems. This review provides an overview of the transition metal complexes used in photopolymerization reactions, with particular emphasis on recent applications in 3D printing and the challenges in achieving highly efficient and sustainable processes.

Exploring Functional Photonic Devices made from a Chiral Metal-Organic Framework Material by a Multiscale Computational Method

Summary: Electronic circular dichroism is an important optical phenomenon for studying chiral molecular materials. Metal-organic frameworks (MOFs) are crystalline porous thin-film materials with tailored chemical and physical properties. Combining these two aspects, researchers aim to create devices with unprecedented functionality. However, advanced modeling techniques are needed to address the complexity of material choices and structural details. A computational workflow that combines quantum chemical methods with optical simulations is proposed, focusing on chiral properties in UiO-67-BINOL MOFs.

Metal-Organic Framework Thin Films as Ideal Matrices for Azide Photolysis in Vacuum

Jimin Song, Xiaojuan Yu, Alexei Nefedov, Peter G. Weidler, Sylvian Grosjean, Stefan Braese, Yuemin Wang, Christof Woell

Summary: This study demonstrates a method to observe UV-induced photochemical reactions of aryl azides within a crystalline matrix in vacuum. By using metal-organic frameworks (MOFs) and surface-mounted MOFs (SURMOFs) as model systems, the azide-related chemical processes can be studied under ultrahigh vacuum (UHV) conditions, excluding solvent effects and in a wide temperature range. In situ infrared reflection absorption spectroscopy (IRRAS) was used to monitor the photoreaction of azide in SURMOFs, revealing the reaction mechanism.

Marriage of an N-Fused and a Regular Porphyrin in a Cofacial Ligand System

Christoph Schissler, Erik K. Schneider, Michael Rotter, Silke Notter, Carsten Geier, Patrick Weis, Manfred M. Kappes, Stefan Braese

Summary: We have developed a synthetic route to achieve cofacial stacking of N-fused porphyrins and regular porphyrins using an o-substituted phenylene linker. This approach involves a crucial Suzuki-Miyaura cross-coupling reaction to introduce a phenyl moiety into the tri-pentacyclic ring of the N-fused porphyrin, enabling the formation of a spatial arrangement that can hold both N-fused porphyrin and regular porphyrin in close proximity. The resulting ligand system was obtained with a 0.77% yield in five steps and was characterized using various techniques.
