Degradation of polysaccharides from Sargassum fusiforme using UV/H2O2 and its effects on structural characteristics

The depolymerization effect of UV/H2O2 on the polysaccharides from Sargassum fusiforme (PSF), a brown algae, were studied. The structural changes of PSF before and after UV/H2O2 treatment were analyzed, and molecular weight changes during in vitro digestion were determined. Results indicated that the molecular weight of PSF was reduced from similar to 289 to similar to 12.6 kDa within 2 h with UV/150 mmol/L H2O2, and the depolymerization effect of UV/H2O2 was significantly higher than that of UV or H2O2 alone. In addition, the UV/H2O2 treatment had a high recovery rate of total sugar (93.54 %) and clearance rate of protein (76.34 %). The monosaccharide composition showed that UV/H2O2 treatment could increase the mole percentage of mannose (37.44 %) and decrease the mole percentage of fucose (14.88 %). The helix-coil transition, X-ray diffraction (XRD) and atomic force microscopy (AFM) imaging showed that the UV/H2O2 treatment depolymerized PSF. Rheological studies indicated that PSF with UV/H2O2 treatment had lower viscosity. In vitro digestion showed that PSF was minimally digested with the in vitro gastrointestinal tract simulation, but PSF with UV/H2O2 treatment could be digested in the low acid environment in the simulated gastric juice, but was minimally digested with the simulated intestinal juice. This studied suggested that the preparation and application of functional PSF with low molecular weight might be beneficial.

Combination Effects of Polyphenols Present in Sugarcane on Proliferation in MCF-7 Human Breast Cancer Cells

Influence of phenolic acids/aldehydes on color intensification of cyanidin-3-O-glucoside, the main anthocyanin in sugarcane (Saccharum officinarum L.)

Effects of UV/H2O2 degradation and step gradient ethanol precipitation on Sargassum fusiforme polysaccharides: Physicochemical characterization and protective effects against intestinal epithelial injury

Summary: In this study, a degraded purified fraction from Sargassum fusiforme polysaccharides (SFP), called DSFP, was produced by UV/H2O2 degradation and step gradient ethanol precipitation. DSFP showed improved protective effects against intestinal epithelial injury, promoting wound healing and cell migration. It also enhanced intestinal barrier function and attenuated intestinal inflammation.

Protective Effect of Peptides from Pinctada Martensii Meat on the H2O2-Induced Oxidative Injured HepG2 Cells

Polysaccharides from brown seaweed: Physicochemical properties, absorption in the intestine, and beneficial effects on intestinal barrier

Summary: The intestinal barrier is vital for maintaining gut homeostasis and overall health. Dysfunction of the intestinal barrier is linked to various diseases such as inflammatory bowel disease and colorectal cancer. This review focuses on the physicochemical properties of three main types of brown seaweed polysaccharides (BSP), their absorption and metabolism in the intestine, and their impact on the intestinal barrier. BSP are not directly digested in the upper gastrointestinal tract but can be utilized through fermentation by gut microbiota. They improve intestinal integrity, reduce tight junction dysfunction and intestinal permeability, and promote the production of beneficial metabolites. This review highlights the potential of BSP derived from brown seaweed as a natural and effective approach for preventing and treating intestinal diseases.

Summary: Liver disease is a major cause of global mortality, and identifying biomarkers for diagnosing its progression is crucial for improving outcomes. Targeted mass spectrometry is a powerful tool for verifying biomarker candidates and clinical applications, particularly for glycoproteins translation. However, the limitation of analyzing only one sample per run has become apparent. In this study, a high-throughput intact N-glycopeptides quantification strategy was developed, allowing the validation of 20 samples per run with an average analysis time of 3 minutes per sample. The strategy was applied in a cohort of 461 serum samples and identified a panel of 10 IgG N-glycopeptides that have strong clinical utility in evaluating the severity of liver disease.

Structural elucidation of an active polysaccharide from Radix Puerariae lobatae and its protection against acute alcoholic liver disease
