Attenuative role of mangiferin in oxidative stress-mediated liver dysfunction in arsenic-intoxicated murines

Mangiferin (MAG), a natural xanthone mainly derived from mangoes, possesses great antioxidative potentials. The present study has been carried out to investigate the hepato-protective role of MAG, against arsenic (As)-induced oxidative damages in the murine liver. As, a well-known toxic metalloid, is ubiquitously found in nature and has been reported to affect nearly all the organs of the human body via oxidative impairment. Administration of As in the form of sodium arsenite (NaAsO2) at a dose of 10 mg/kg body weight for 3 months abruptly increased reactive oxygen species (ROS) level, led to oxidative stress and significantly depleted the first line of antioxidant defense system in the body. Moreover, As caused apoptosis in hepatocytes. Treatment with MAG at a dose of 40 mg/kg for body weight for 30 days simultaneously and separately after NaAsO2 administration decreased the ROS production and attenuated the alterations in the activities of all antioxidant indices. MAG also protected liver against the NaAsO2-induced apoptosis and disintegrated hepatocytes, thus counteracting with As-induced toxicity. It could significantly inhibit the expression of different proapoptotic caspases and upregulate the expression of survival molecules such as Akt and Nrf2. On inhibiting Akt (by PI3K inhibitor, LY294002) and ERK1/2 (by ERK1/2 inhibitor, PD98059) specifically, caspase 3 got activated abolishing mangiferin's protective role on As-induced hepatotoxicity. So here, we have briefly elucidated the signaling cascades involved in As-induced apoptotic cell death in the liver and also the detailed cellular mechanism by which MAG provides protection to this organ. Melatonin counteracts necroptosis and pulmonary edema in cadmium-induced chronic lung injury through the inhibition of angiotensin II

Summary: The study demonstrates the efficacy of melatonin in controlling the renin-angiotensin system (RAS) in the context of chronic lung injury induced by cadmium (Cd) exposure. Melatonin treatment effectively inhibits inflammatory responses and necroptotic cell death mediated by Angiotensin-II (Ang-II), thereby protecting the alveolar-capillary barrier from Cd toxicity.

pH-responsive and targeted delivery of rutin for breast cancer therapy via folic acid-functionalized carbon dots

Summary: In this study, carbon dots (CDs) were synthesized from red Korean ginseng and further modified with folic acid (FA) to deliver rutin (RUT) for targeted cancer therapy. The resulting FA conjugated and rutin loaded CDs (FA-CDs-RUT) exhibited size of approximately 70 nm, drug loading content of 14.5%, and a pH-dependent drug release behavior. The nanohybrid demonstrated dose-dependent cytotoxicity and anti-migratory effect in human breast cancer MCF-7 cells. In vivo administration of FA-CDs-RUT was found to effectively inhibit tumor growth in mice without causing significant alterations in blood biochemical parameters. Molecular docking study revealed the potential interaction between rutin and Bcl-2, suggesting a possible mechanism of action. Overall, this study highlights the potential of FA-CDs-RUT for breast cancer treatment.

Folic acid-functionalized cerium oxide nanoparticles as smart nanocarrier for pH-responsive and targeted delivery of Morin in breast cancer therapy

Summary: In this study, folic acid-conjugated and amine-functionalized cerium oxide nanoparticles (CeO2-NH-FA) were synthesized for targeted and controlled delivery of Morin to breast cancer cells overexpressing folic acid. The resulting nanohybrids exhibited significant cytotoxicity against breast cancer cells and a potential antimigratory effect. In addition, the nanohybrids showed improved anticancer efficacy compared to free Morin and CeO2-NH-FA in tumor-bearing mice. The study highlights the potential of the Morin-CeO2-NH-FA nanohybrids as a drug delivery vehicle for cancer treatment.

The role of macrophages in the tumor microenvironment and tumor metabolism

Summary: The complexity and plasticity of the tumor microenvironment make it challenging to comprehend the intratumoral regulation of different cell types and their activities. Tumor-associated macrophages (TAMs), a subtype of macrophages, have a significant impact on the signaling dynamics of the tumor microenvironment. TAMs play a crucial role in regulating cancer cell activities, immunosuppression, and resistance to chemotherapy and immunotherapy. This review focuses on the mechanisms by which TAMs modulate immune signaling in the tumor microenvironment and recent preclinical research on developing therapeutics targeting TAMs and tumor metabolism.

Summary: Selective initiation of programmed cell death in cancer cells was achieved by a synthetic bis-coumarin derivative 3 l, which demonstrated promising cytotoxic ability and apoptosis induction specifically in human renal carcinoma cells (SKRC-45). This compound modulated apoptotic pathways and inhibited NF-kappa B nuclear localization, resulting in reduced proliferation, survival, and migration ability of SKRC-45 cells. In an in vivo tumor model, 3 l exhibited an anticancer effect without affecting kidney function.
