Summary: Neuroendocrine prostate cancer (NEPC) is an aggressive subtype characterized by loss of androgen receptor signaling and transdifferentiation toward small-cell neuroendocrine phenotypes. Using SCN phenotype scores and gene depletion screens, ZBTB7A was identified as a candidate promoting NEPC progression. High SCN phenotype scores correlated with reliance on RET kinase activity and dependence on ZBTB7A. ZBTB7A showed distinct gene networking patterns in NEPC compared to prostate adenocarcinoma, with a strong association with genes promoting cell cycle progression. Silencing ZBTB7A suppressed cell growth and induced apoptosis in NEPC cell lines.

Summary: This study analyzed the RNA-sequencing and whole-exome sequencing data of 170 patients with neuroendocrine prostate cancer (NEPC). It found that NEPC has a unique tumor immune landscape compared to other prostate cancer types and small-cell lung cancer (SCLC). NEPC is characterized by a relatively immune-depleted tumor immune microenvironment and less mutations, but has comparable expression of checkpoint genes PD-L1 and CTLA-4 with SCLC. These findings can inform the development of immunotherapy strategies for NEPC.

Summary: Prostate cancer is the most common malignancy in men worldwide. Cancer stem cells, which possess unique properties and are responsible for therapy resistance and disease relapse in several malignancies including prostate cancer, can be identified by certain markers such as ALDH EZH2, OCT4, SOX2, c-MYC, and Nanog. Advances in the field offer explanations for uncertainties surrounding the etiology and hope for the development of new stem-cell targets and efficient therapies in the future. This review discusses the identification, properties, pathways, diagnostics, and interventions of prostate cancer stem cells.
