Drug resistance remains a severe problem in most chemotherapy regimes. Recently, it has been suggested that cancer cell-derived extracellular vesicles (EVs) could mediate drug resistance. In this study, the role of EVs in mediating the response of oral squamous cell carcinoma (OSCC) cells to cisplatin was investigated. We isolated and characterized EVs from OSCC cell lines showing differential sensitivities to cisplatin. Increased EV production was observed in both de novo (H314) and adaptive (H103/cisD2) resistant lines compared to sensitive H103 cells. The protein profiles of these EVs were then analyzed. Differences in the proteome of EVs secreted by H103 and H103/cisD2 indicated that adaptation to cisplatin treatment caused significant changes in the secreted nanovesicles. Intriguingly, both resistant H103/cisD2 and H314 cells shared a highly similar EV protein profile including downregulation of the metal ion transporter, ATP1B3, in the EVs implicating altered drug delivery. ICP-MS analysis revealed that less cisplatin accumulated in the resistant cells, but higher levels were detected in their EVs. Therefore, we inhibited EV secretion from the cells using a proton pump inhibitor and observed an increased drug sensitivity in cisplatin-resistant H314 cells. This finding suggests that control of EV secretion could be a potential strategy to enhance the efficacy of cancer treatment.

Application of chitosan-based nanoparticles in skin wound healing

Hooi Leong Loo, Bey Hing Goh, Learn-Han Lee, Lay Hong Chuah

Summary: With the increasing prevalence of impaired wound healing, there has been a growing interest in the development of novel wound dressings with enhanced healing functions. Nanoparticle-based delivery systems, particularly chitosan-based nanoparticles, have emerged as promising candidates due to their non-toxicity, biocompatibility, and bioactivity. This review focuses on the application of chitosan-based nanoparticle systems in wound treatment, discussing their properties, mechanisms, and potential for improving wound healing. The results show that chitosan-based nanoparticles exhibit superior wound healing effects, either alone or as drug delivery vehicles. Overall, there are great opportunities and potentials for the use of chitosan nanoparticles in wound healing.

Bacterial extracellular vesicles in biofluids as potential diagnostic biomarkers

Kar-Yan Su, Jie-Yi Koh Kok, Yie-Wei Chua, Shearn-Dior Ong, Hooi Leng Ser, Priyia Pusparajah, Pui San Saw, Bey Hing Goh, Wai-Leng Lee

Summary: Extracellular vesicles (EVs) are membrane-derived lipid bilayers released by cells. Bacterial EVs (BEVs) have attracted interest as potential diagnostic markers due to their differential bacterial composition in biofluids between patients and healthy controls. This review outlines the commonly used approach for investigating BEVs in biofluids, including 16S ribosomal RNA gene sequencing, and discusses the potential of BEVs as biomarkers for various diseases.

The Convergence of FTIR and EVs: Emergence Strategy for Non-Invasive Cancer Markers Discovery

Le-Wei Wong, Siow-Hui Mak, Bey-Hing Goh, Wai-Leng Lee

Summary: In conjunction with imaging analysis, pathology-based assessments of biopsied tissue are the gold standard for diagnosing solid tumors. However, the disadvantages of tissue biopsies have led to the development of liquid biopsy, which involves sampling and clinical assessment of bodily fluids for cancer diagnosis. Extracellular vesicles (EVs), carrying molecular profiles of their cell or tissue origins, have emerged as one of the most promising biomarkers for cancer, allowing for non-invasive and rapid diagnosis with the help of Fourier-transform infrared spectroscopy (FTIR).

Expression of Epsin3 and its interaction with Notch signalling in oral epithelial dysplasia and oral squamous cell carcinoma

Halah Ahmed, Ian Paterson, Sadat A. A. Aziz, Ottavio Cremona, Max Robinson, Marco Carrozzo, Ruth A. Valentine

Summary: In this study, the role of Epsin3 and Notch1 in oral carcinogenesis was investigated, aiming to explore their potential as biomarkers. The results showed that Epsin3 was upregulated in oral epithelial dysplasia and oral squamous cell carcinoma, suggesting its potential as a biomarker for oral epithelial dysplasia. Notch signaling was downregulated in oral squamous cell carcinoma, possibly through an Epsin3-induced de-activation pathway.

Summary: This study examined the antibacterial effects of Mentha longifolia essential oil and pulegone in edible coatings on the growth of Staphylococcus aureus, Listeria monocytogenes, and Escherichia coli in cheese. The results showed that the addition of these compounds significantly reduced bacterial growth during storage. Increasing the concentration of chitosan, M. longifolia, and pulegone in the coatings further decreased the bacterial population, with pulegone exhibiting a stronger effect. Overall, the findings suggest that M. longifolia essential oil and pulegone, combined with alginate and chitosan coatings, have antibacterial effects against S. aureus, L. monocytogenes, and E. coli in cheese.

Summary: This study aimed to identify and review research related to the practice of patients with DM in Brunei Darussalam in order to identify associated factors influencing diabetic medication knowledge and adherence. The results revealed relatively poor medication knowledge and inappropriate diabetes-related practices in studies conducted in Brunei Darussalam. However, no study has directly measured both medication awareness and compliance among Brunei Darussalam T2DM patients. Future studies can focus on the relation between these two factors in different geographical locations characterized by different diabetes-related practices and culture.
