Background: To help inform decision making in the clinical setting, we carried out a systematic review and meta-analysis to estimate the association of thyroid disease risks with obesity. Methods: Pubmed, Embase, Web of Science, Cochrane database and Google Scholar electronic databases were searched from inception to October 31, 2018 without language restrictions to explore the relationship between thyroid disorders and obesity. The relative risk (RR) or odds risk (OR) for thyroid disorders were pooled using the SPSS and STATA software. Results: A total of 22 studies were included in the study. (1) Meta-analysis showed that obesity was significantly associated with an increased risk of hypothyroidism (RR = 1.86, 95% CI 1.63-2.11, P < 0.001). Meta-analyses after stratification further showed that obese population had increased risks of overt hypothyroidism (RR = 3.21, 95% CI 2.12-4.86, P < 0.001) and subclinical hypothyroidism (RR = 1.70, 95% CI 1.42-2.03, P < 0.001). (2) Further meta-analysis also showed obesity was clearly associated with Hashimoto's thyroiditis (RR = 1.91, 95% CI 1.10-3.32, P = 0.022), but not with Graves' disease. (3) In themeta-analysis of antibodies, obesity was correlated with positive thyroid peroxidase antibody (TPOAb) (RR = 1.93, 95% CI 1.31-2.85, P = 0.001), but not with positive thyroglobulin antibody (TGAb). Conclusions: Obesity was significantly related to hypothyroidism, HT, and TPOAb, implying that prevention of obesity is crucial for thyroid disorders.

Summary: The study found that the SNP rs3778638 in the PSORS1C1 gene is associated with susceptibility to Graves' disease, but not with Hashimoto's thyroiditis. Additionally, no linkage disequilibrium was observed for the other three SNPs in PSORS1C1 among patients with AITD and controls.

Summary: The study identified exosomal candidate proteins in the pathogenesis of Rheumatoid Arthritis (RA) among which 32 were upregulated and 5 were downregulated. These proteins interacted with each other and were mainly involved in inflammatory-related pathways, suggesting their role in mediating the onset of RA.

Polymorphisms of ATG5 Gene Are Associated with Autoimmune Thyroid Diseases, Especially Thyroid Eye Disease

Wen Wang, Zheng-yao Yu, Rong-hua Song, Shuang-tao He, Liang-feng Shi, Jin-an Zhang

Summary: This study explores the association between ATG5 gene polymorphisms and AITDs, specifically HT and GD. The research finds that allele A frequency of rs6568431 and allele G frequency of rs6937876 are significantly higher in AITDs patients, while the G allele frequency of rs6937876 is significantly higher in GD patients with ophthalmopathy. This study reveals the association between ATG5 gene and AITDs for the first time.

Inclusion of ALKBH5 as a candidate gene for the susceptibility of autoimmune thyroid disease

Summary: This study investigated the association between ALKBH5 gene polymorphisms and autoimmune thyroid disease (AITD), focusing on the variants rs9913266 and rs12936694 which showed significant differences in AITD and Graves' disease. Moreover, the minor allele of rs9913266 was found to be associated with the risk of female AITD and Hashimoto's thyroiditis development. Additionally, haplotypes CGACA and CAGCG were significantly correlated with susceptibility to AITD and Graves' disease.

METTL3gene polymorphisms contribute to susceptibility to autoimmune thyroid disease

Rong-hua Song, Xue-rong Liu, Chao-qun Gao, Peng Du, Jin-an Zhang

Summary: The study did not find a significant correlation between METTL3 genotypes at seven SNPs and AITD development among both GD and HT patients, except for a borderline association between rs3752411 and GD under certain conditions. Subgroup analysis showed that certain SNPs had higher minor allele frequencies in male AITD patients compared to healthy volunteers, while the genotype distribution of another SNP was significantly different between the groups. Multivariate logistic regression analyses revealed a significant correlation between specific allele frequencies and GD in male patients, indicating a potential role of METTL3 as a candidate gene for AITD treatment.
