Summary: Increasing evidence suggests that neuroinflammation may be a contributing factor to the failure of clinical antidepressants. The NLRP3 inflammasome, a multiprotein complex, has been identified as a key player in the development of neuroinflammation. Inhibition of NLRP3 inflammasome activation presents a promising therapeutic strategy for depression. This review summarizes current research on the role of NLRP3 inflammasome in depression pathology, including its activation in patients and animal models, potential driving mechanisms, and its pathogenetic role in depression. Additionally, it provides an overview of current and potential antidepressants targeting NLRP3 inflammasome.

Summary: In this study, the mechanisms of ACR-induced neuroinflammation and pyroptosis were investigated using a neuron cell model. The results showed that ACR treatment induced lytic cell death through two different pyroptotic pathways in SH-SY5Y cells. Additionally, the NLRP3 inflammasome cascade was found to be activated first.

Summary: The NLRP3 inflammasome is activated through mitochondria-mediated ubiquitination by the E3 ligase MARCH5. Knockout of MARCH5 leads to an impaired secretion of IL-1β and IL-18 in myeloid cells, as well as a decreased mortality rate upon infection. MARCH5-dependent NLRP3 ubiquitination is essential for NLRP3-NEK7 complex formation and NLRP3 oligomerization.

Summary: In this study, the protective effect of CX3CL1 against cerebral ischemia was investigated. It was found that exogenous rCX3CL1 administration after the ischemic insult exerted a long-term neuroprotective effect on post-ischemic stroke by inhibiting NLRP3 inflammasome-induced microglial pyroptosis. These findings suggest that the CX3CL1 signaling pathway can serve as a therapeutic target for promoting the functional recovery after stroke.

Summary: This study found that the formation of the NEK7-NLRP3 complex significantly increased during S. pneumoniae infection and ALK and JNK were rapidly phosphorylated. Inhibitors of ALK and JNK reduced the ability of bacterial killing, the gene expression of NLRP3 inflammasome, the formation of ASC specks and the NEK7-NLRP3 complex, leading to decreased activation of NLRP3 inflammasome-associated molecules and maturation of IL-1β. Interestingly, the ALK/JNK/NEK7-NLRP3 signaling pathway also regulated pyroptosis and IL-1β secretion triggered by S. pneumoniae infection.

Summary: The membrane-bound E3 ubiquitin ligase gp78 mediates mixed ubiquitination of NLRP3, restraining inflammasome activation by inhibiting NLRP3 oligomerization and subcellular translocation. Insig-1, an endoplasmic reticulum membrane protein, is required for the gp78-NLRP3 interaction and gp78-mediated NLRP3 ubiquitination. Deficiency of gp78 or Insig-1 in myeloid cells leads to exacerbated NLRP3 inflammasome-dependent inflammation in vivo.
