Summary: The successful establishment of endometrial receptivity is crucial for the fertility and economic benefits of ewes. In this study, high-fertility Hu sheep were found to have higher uterine gland density, uterine coefficient, and number of uterine caruncles compared to low-fertility Hu sheep during the luteal phase. RNA sequencing (RNA-Seq) analysis identified thousands of differentially expressed genes in the endometrium of Hu sheep with different fertility potential. One of these genes, CREB1, was found to be differentially expressed in the Hu sheep endometrium during the estrous cycle. Functional analysis revealed that CREB1 significantly influenced the expression level of endometrial receptivity marker genes, promoted cell proliferation, and inhibited cell apoptosis and autophagy. Additionally, there was a negative linear correlation between miR-134-5p and CREB1 in the endometrium, and overexpression of CREB1 counteracted the negative effect of miR-134-5p on endometrial stromal cell growth.

Summary: In this study, the clinical implications of actin filament associated protein 1-antisense RNA 1 (AFAP1-AS1) in endometriosis (EMS) were investigated. It was found that high expression of AFAP1-AS1 and BCL9, as well as low expression of miR-15a-5p, were observed in EMS. Exosomes derived from endometrial stromal cells (ESCs) were found to deliver AFAP1-AS1 to ESCs, promoting their proliferation, migration, and invasion. AFAP1-AS1 bound to BCL9, which was targeted by miR-15a-5p in EMS. Inhibition of exosomal AFAP1-AS1 suppressed migration and invasion of ESCs through miR-15a-5p/BCL9. These findings suggest that ESCs-derived exosomes carrying AFAP1-AS1 contribute to the pathogenesis of EMS.
