A novel isoquinoline compound abolishes chronic unpredictable mild stress-induced depressive-like behavior in mice

Chronic unpredictable mild stress (CUMS) elicits aspects of cognitive and behavioral alterations that can be used to model comparable aspects of depression in humans. The aim of the present study was to investigate the antidepressant-like potential of 7-fluoro-1,3-diphenylisoquinoline-1-amine (FDPI), a novel isoquinoline compound, in CUMS, a model that meets face, construct and predictive criteria for validity. Swiss mice were subjected to different stress paradigms daily for a period of 35 days to induce the depressive-like behavior. The animals received concomitant FDPI (0.1 and 1 mg/kg, intragastric) or paroxetine (8 mg/kg, intraperitoneal) and CUMS. The behavioral tests (splash test, tail suspension test, modified forced swimming test and locomotor activity) were performed. The levels of cytokines, corticosterone and adrenocorticotropic (ACTH) hormones were determined in the mouse prefrontal cortex and serum. The synaptosomal [H-3] serotonin (5-HT) uptake, nuclear factor (NF)-kappa B, tyrosine kinase receptor (TrkB) and pro-brain-derived neurotrophic factor (BDNF) levels were determined in the mouse prefrontal cortex. CUMS induced a depressive-like behavior in mice, which was demonstrated in the modified forced swimming, tail suspension and splash tests. FDPI at both doses prevented depressive-like behavior induced by CUMS, without altering the locomotor activity of mice. FDPI at the highest dose prevented the increase in the levels of NF-kB, pro-inflammatory cytokines, corticosterone and ACTH and modulated [H-3]5-HT uptake and the proBDNF/TrkB signaling pathway altered by CUMS. The present findings demonstrated that FDPI elicited an antidepressant-like effect in a model of stress-induced depression. (C) 2016 Elsevier B.V. All rights reserved.

m-CF3-substituted diphenyl diselenide attenuates all phases of morphine-induced behavioral locomotor sensitization in mice

Summary: The study found that (mCF3-PhSe)(2) attenuated morphine-induced locomotor sensitization by modulating weight gain, locomotor activity, and protein contents of opioid and glutamate receptors in the cerebral cortex. The compound did not affect dopaminergic receptors in the acquisition phase, but was effective against behavioral and molecular alterations caused by morphine at all phases of locomotor sensitization.

The compound showed penetration into the central nervous system and low toxicity at a high dose, making it an interesting candidate for the development of a new therapeutic strategy for major depressive disorder (MDD).

Summary: The study found that the functionalized 3-selanyl benzo[b]furan (SeBZF) has antidepressant-like potential in mice. Specifically, SeBZF3 exhibited antidepressant-like effects and interacted with 5-HT1A, 5-HT2A/C, and 5-HT3 receptors. Furthermore, SeBZF3 showed synergistic action with the selective serotonin reuptake inhibitor fluoxetine and did not induce acute oral toxicity at high doses.

Summary: This study investigated the anxiolytic-like effect of (m-CF3-PhSe)2 and its modulation of synaptic plasticity and NMDAR-mediated neurotoxicity in young mice exposed to a lifestyle model. The results showed that (m-CF3-PhSe)2 abolished anxiety-like behavior, reversed neurotoxicity, and restored synaptic plasticity-related signaling. These findings suggest that (m-CF3-PhSe)2 may be a potential candidate for anxiolytic drug development.

Summary: In this study, the efficacy of (m-CF3-PhSe)2 against lifestyle model-related metabolic disturbances and hypothalamic inflammation in young mice was investigated. The results showed that (m-CF3-PhSe)2 reduced relative adipose tissue weight, hyperglycemia, and dyslipidemia in mice exposed to the lifestyle-induced model. It also modulated various metabolic and inflammatory markers in the liver and hypothalamus of the mice.

Summary: Mood disorders caused by chemotherapy have become more important as cancer patients' survival increases. This study used methotrexate to induce mood disorders in rats and found that treatment with the antioxidant N-acetylcysteine (NAC) can alleviate anxiety and depression-like behaviors, increase antioxidant capacity, reduce oxidative stress and inflammatory response, and regulate brain chemistry. The findings suggest that NAC treatment could be an effective strategy in revising the treatment for individuals suffering from chemotherapy-induced mood disorders.

Continuous high-frequency repetitive transcranial magnetic stimulation at extremely low intensity affects exploratory behavior and spatial cognition in mice

Clinical effects of anodal tDCS and identifying response markers in post-traumatic stress disorder (PTSD): An open-label study

Self-esteem and cortical thickness correlate with aggression in healthy children: A surface-based analysis

Summary: Aggression can have serious consequences, but little is known about its personality and neurological origins in children. This study investigated the relationship between self-esteem, aggression, and brain structure in healthy children, and found that self-esteem was negatively associated with aggression. The study also revealed that increased cortical thickness in certain brain areas may be a potential mechanism linking low self-esteem to aggression in children.

Role of serotonin in the lack of sensitization caused by prolonged food deprivation in Aplysia

Summary: Food deprivation can lead to neurological dysfunctions, including memory impairment. This study used Aplysia as an animal model to investigate the memory deficits caused by prolonged food deprivation. The results showed that 14 days of food deprivation decreased the level of 5-HT in the hemolymph, which contributed to the lack of sensitization and its cellular correlates. However, exogenous application of 5-HT partially induced sensitization in the food deprived animals.

The effects of a dual orexin receptor antagonist on fear extinction memory and sleep in mice: Implications for exposure therapy

Summary: The study found that intervention with the dual orexin receptor antagonist suvorexant did not have the expected effects on extinction memory and sleep. Higher percentages of REM sleep were associated with poorer extinction memory recall and stronger fear responses. Coordination function index: A novel indicator for assessing hindlimb locomotor recovery in spinal cord injury rats based on catwalk gait parameters

Transcranial alternating current stimulation does not affect microscale learning
