Increased levels of proapoptotic markers in normal ovarian cortex surrounding small endometriotic cysts

Endometriosis can impair fertility by reducing ovarian reserve and the production of good-quality oocytes. The surgical removal of endometriotic lesions is generally recommended for women who wish to conceive. In this paper we studied whether ovarian cortex adjacent to excised small (diameter <= 4 cm) endometriotic cyst (here referred as Cortex Surrounding Endometriotic Cyst, CSEC) showed signs of tissue damages by evaluating the expression of proteins involved in DNA repair and apoptosis. To this end, phosphorylated H2A.X, Chk1 and 2, ATM and ATR, Bcl-2, Bid, phosphorylated and total p53, caspases (9, 8 and 3), XIAP, phosphorylated and total NF kappa B were analyzed by western blot. Results showed that caspase 8, XIAP, p53/p-p53 and NF.B were more abundantly expressed in all samples of CSEC group in comparison with ovarian cortex of controls. Conversely, the levels of the other proteins were comparable between the two groups. In conclusion, these results suggest that NF.B, caspase 8 and p53/p-p53 elevated expressions in samples of CSEC can be considered as an early sign of tissue injury, indicating that ovarian cortex is already sensitized to apoptosis and inflammation. Therefore, excision of EC should occur very early, to avoid further ovarian damages.

Recent advances have explored the molecular mechanisms and signaling pathways that regulate mitophagy, including the early role and spatial specificity of the AMPK/ULK1 axis.

However, fluconazole shows no cytotoxic effects.

Summary: This study investigated the inhibitory activity of protocetraric and salazinic acids against SARS-CoV-2 3CL(pro). The results showed that both compounds are slow-binding inactivators of 3CL(pro) with a competitive inhibition mechanism. In cellular experiments, these compounds did not show cytotoxicity. Molecular modeling predicted the binding mechanism of these compounds with 3CL(pro).

Summary: This study evaluated the in vitro effects of preservative-free bimatoprost (BIM) 0.03% vs. 0.01% on human conjunctival epithelial (HCE) cells. The results showed that long-term exposure to BIM 0.03% resulted in a significant decrease in cell proliferation and viability, as well as alterations in cell cycle and mitochondrial function. BIM 0.01% did not exhibit cytotoxicity or negative influence on conjunctival cell growth and viability.

Endocannabinoid System Components of the Female Mouse Reproductive Tract Are Modulated during Reproductive Aging

Summary: This study aimed to investigate the expression levels of the main receptors and metabolic enzymes of the endocannabinoid system in the ovaries, oviducts, and uteri of mice at different reproductive stages. The results showed that TRPV1 receptor had the highest expression and significantly increased during aging, while NAPE-PLD, FAAH, and DAGL-beta enzymes were the most expressed in these organs and increased with age. Immunohistochemistry revealed the distribution of these proteins in specific cells in each organ. These findings provide new insights into the role of the endocannabinoid system in female reproduction and its therapeutic potential.

Summary: Controlled ovarian stimulation (COS) is a common procedure in assisted reproductive technologies, but it can lead to an unbalanced hormonal and molecular environment that affects cellular mechanisms. This study found changes in mitochondrial DNA fragmentation, antioxidant enzymes, apoptotic proteins, and pro-survival proteins in the oviducts of mice that underwent repeated hyperstimulation. The results show that while the antioxidant machinery is activated, apoptosis is not induced due to the activation of pro-survival proteins.

Do Aging and Parity Affect VEGF-A/VEGFR Content and Signaling in the Ovary?-A Mouse Model Study
