Generating amplicon reads for microbial community assessment with next-generation sequencing

Marker gene amplicon sequencing is often preferred over whole genome sequencing for microbial community characterization, due to its lower cost while still enabling assessment of uncultivable organisms. This technique involves many experimental steps, each of which can be a source of errors and bias. We present an up-to-date overview of the whole experimental pipeline, from sampling to sequencing reads, and give information allowing for informed choices at each step of both planning and execution of a microbial community assessment study. When applicable, we also suggest ways of avoiding inherent pitfalls in amplicon sequencing.

Sea beet's endophytic microbiome is more diverse and salt resistant than its crop relative, and supplementing trehalose during lyophilization is more effective in enhancing bacterial community salt stress tolerance, viability, and density. This suggests the potential use of trehalose in formulating improved biofertilizers.

A Combined Analysis of Gut and Skin Microbiota in Infants with Food Allergy and Atopic Dermatitis: A Pilot Study

Summary: This study found a relationship between gut and skin microbiota in patients with allergies, especially in terms of bacterial community structure and allergy phenotypes. Different clinical conditions were associated with characteristic bacterial taxa in the skin and gut microbiota, and while infants with allergies and healthy infants showed similar alpha-diversity in their microbiota, differences in beta-diversity and bacterial species abundance were observed. Positive correlations were found between certain bacterial taxa in skin and fecal samples, which may serve as indicators of the host's allergic state.

Summary: Recent studies have highlighted the crucial role of post-transcriptional processes in gene expression regulation, particularly the retention of mRNA in the nucleus. Analysis of larch microsporocytes revealed two main patterns of nuclear accumulation and transport of transcripts, indicating an evolutionarily conserved mechanism of gene regulation across various cell types.

MicroRNA as a Potential Therapeutic Molecule in Cancer

Summary: miRNAs are not only markers of neoplastic disease initiation and progression, but also markers of response to anticancer therapy. Several miRNAs have been identified as biomarkers of drug resistance and have potential to sensitize cancer cells to therapy. The use of miRNAs in cancer treatment has been verified in clinical trials, showing great potential.

Summary: Breast cancer demonstrates diverse epigenetic abnormalities that impact gene expression and tumor characteristics. Epigenetic alterations are crucial in cancer development and progression, and drugs targeting epigenetic modifications show promise for cancer treatment, including DNA methyltransferase inhibitors, histone-modifying enzymes, and mRNA regulators. However, currently, there is no effective standalone epigenetic drug therapy for breast cancer. Combining epigenetic drugs with conventional therapies has shown positive results, and targeting specific epigenetic changes may lead to more effective monotherapy options in the future.
