Summary: Excessive fluoride exposure can lead to pathological changes in testicular tissues, affecting oxidative stress, essential elements levels, and DNA damage. SOD1 activity and DNA damage are the most sensitive indicators, with the lowest BMDLs at 0.1 µg F-/kg b.w. This study provides valuable insights for further research on the health risks associated with fluoride exposure in both human and animal populations.

Summary: The study aimed to investigate the effects of BPA on DNA damage and cytotoxicity in HepG2 cell line at concentrations ranging from 0.02-200μg/mL, as well as the ameliorating effect of GA (10-50μg/mL). The results showed that BPA was not genotoxic and cytotoxic at low dose exposure (0.02μg/mL), but induced significant DNA damage and cytotoxicity in a dose-dependent manner at concentrations of 2-200μg/mL. However, the addition of GA (10-50μg/mL) with BPA (200μg/mL) inhibited DNA damage and cytotoxicity.

Summary: This study generated a DNA methylome and transcriptome atlas of precocious porcine testicular tissues, providing insights into the dynamic regulatory mechanisms of DNA methylation during testicular development. The results showed a direct relationship between dynamic methylation patterns and the expression of the DNMT3A gene. Promoter methylation was found to negatively regulate 3' UTR methylation. Additionally, decreased promoter methylation affected the upregulation of meiosis-related genes, while demethylation in the 3' UTR led to the downregulation of the INHBA gene. These findings contribute to a better understanding of the regulatory mechanisms of DNA methylation in testicular development.

Summary: DNA damage is a common cellular feature in cancer and neurodegenerative disease, but there is a lack of fast and accurate methods for quantifying DNA damage. The comet assay is a biochemical tool that measures DNA damage based on the migration of broken DNA strands towards a positive electrode, creating a quantifiable 'tail' behind the cell. However, the analysis of comets using existing open-source algorithms is time-consuming and requires manual curation. To overcome these limitations, we developed AutoComet, an open-source algorithm that automates comet segmentation and quantification of tail parameters. AutoComet significantly improves segmentation accuracy and reduces analysis time over ten-fold, providing a fast and unbiased method for quantifying DNA damage.

Summary: A new material composed of kaolin base with silver nanoparticles was developed as an alternative to antibiotics in animal feed. In an in vivo geno-toxicological evaluation in rats, the material showed no adverse effects when administered orally at a dose of 2000 mg/kg b.w. for 28 days. The material did not induce micronuclei or DNA damage in various tissues, and silver content in target organs confirmed the negative results.

Summary: The objective of this study was to identify different transcriptome expression profiles involved in the pathogenesis of diabetic nephropathy (DN) and to illustrate the diagnostic and therapeutic potential of mRNAs, lncRNAs, and circRNAs in DN progression. Differential expression of mRNAs, lncRNAs, and circRNAs were found to play important roles in the development of DN. Several potential biomarkers were identified for the diagnosis of DN. The results of this study have significant implications for further research and treatment of this disease.

Summary: The study found that BPB and BPAF, analogs of BPA, have adverse effects on uterine health, causing endometrial contraction, decreased epithelial height, and increased gland numbers. Bioinformatics analysis revealed that both BPB and BPAF disrupt the immune landscape of the uterus. Furthermore, eight genes associated with BPB and BPAF exposure were found to be related to uterine corpus endometrial carcinoma (UCEC), and high expression of Srd5a1 was significantly correlated with poor prognosis in patients.

Summary: In this study, neonatal male mice were exposed to BPA to investigate its effects on testicular development. The results showed that BPA exposure induced apoptosis of testicular cells and reduced the thickness of seminiferous epithelium. BPA also decreased the antioxidant capacity of testes and levels of Nrf2 and its downstream antioxidant molecules. Additionally, BPA exposure resulted in increased m6A modification of Nrf2 mRNA, indicating the involvement of inhibited FTO in BPA-induced testicular oxidative stress and developmental injury.
