Permutation inference methods for multivariate meta-analysis

Multivariate meta-analysis is gaining prominence in evidence synthesis research because it enables simultaneous synthesis of multiple correlated outcome data, and random-effects models have generally been used for addressing between-studies heterogeneities. However, coverage probabilities of confidence regions or intervals for standard inference methods for random-effects models (eg, restricted maximum likelihood estimation) cannot retain their nominal confidence levels in general, especially when the number of synthesized studies is small because their validities depend on large sample approximations. In this article, we provide permutation-based inference methods that enable exact joint inferences for average outcome measures without large sample approximations. We also provide accurate marginal inference methods under general settings of multivariate meta-analyses. We propose effective approaches for permutation inferences using optimal weighting based on the efficient score statistic. The effectiveness of the proposed methods is illustrated via applications to bivariate meta-analyses of diagnostic accuracy studies for airway eosinophilia in asthma and a network meta-analysis for antihypertensive drugs on incident diabetes, as well as through simulation experiments. In numerical evaluations performed via simulations, our methods generally provided accurate confidence regions or intervals under a broad range of settings, whereas the current standard inference methods exhibited serious undercoverage properties.

Diverse and divergent functions of IL-32β and IL-32γ isoforms in the regulation of malignant pleural mesothelioma cell growth and the production of VEGF-A and CXCL8

Summary: This study aimed to investigate the effects of interleukin (IL)-32 beta and IL-32 gamma on mesothelioma cell growth, as well as the expression of vascular endothelial growth factor (VEGF)-A and C-X-C motif chemokine ligand 8 (CXCL8). IL-32 had a growth-promoting effect on one of the six mesothelioma cell lines and had diverse regulatory functions in VEGF-A and CXCL8 secretion from mesothelioma cells stimulated with or without IL-17A. Overexpressing IL-32 gamma in mesothelioma cells resulted in enhanced IL-32 beta expression, affecting cell growth and VEGF-A/CXCL8 expression. IL-32 beta-mediated growth and VEGF-A/CXCL8 production were mainly regulated by the phosphatidylinositol-3 kinase (PI3K) signaling pathway, while IL-32 beta-mediated growth inhibition was regulated by the mitogen-activated protein kinase (MAPK) pathway. In a xenograft model, tumors with IL-32 gamma grew faster and showed increased vascularity compared to control tumors.

Bias in Odds Ratios From Logistic Regression Methods With Sparse Data Sets

Summary: Logistic regression models can lead to bias in sparse data, but Bayesian methods can reduce the bias and are suitable for sparse data analysis. Simulation and real data analysis show that the hyper-g prior modeling and log F-type priors can reduce the bias, but the choice of method depends on the null and alternative hypotheses.

The Global Alliance for Living Evidence on aNxiety, depressiOn and pSychosis (GALENOS) aims to catalog and evaluate relevant scientific research, involving both human and preclinical studies, to advance mental health science. GALENOS will also facilitate collaboration and identify key research questions, accelerating the translation of discovery science into effective interventions.

Diagnostic accuracy of point-of-care ultrasound for shock: a systematic review and meta-analysis

Prevalence of gambling disorder and its correlates among homeless men in Osaka city, Japan

Summary: The prevalence of gambling disorder is higher among homeless individuals than the general population, but the factors associated with gambling disorder in this population are not well understood. This study aimed to investigate the prevalence and associated factors of gambling disorder among homeless men in Osaka City. The results showed that the lifetime and past-year prevalence of gambling disorder were higher among homeless men compared to the general population. Factors associated with lifetime gambling disorder included longer duration of homelessness and more frequent experiences of homelessness.
