Perylene Diimide and Luminol as Potential-Resolved Electrochemiluminescence Nanoprobes for Dual Targets Immunoassay at Low Potential

In the field of clinical diagnosis, it is important to construct a potential-resolved multiplex electrochemiluminescence (ECL) biosensor for decreasing the false-positive rate and improving the diagnostic accuracy. However, the shortage of low-potential cathodic luminophores between -1 and 0 V (vs Ag/AgCl) severely limited the development of the biosensor. Herein, we synthesized a novel luminophore N,N-bis-(3-dimethyl aminopropyl)-3,4,9,10-perylene tetracarboxylic acid diimide (PDI), which gave dual emissions at -0.25/-0.26 V with K2S2O8 as a co-reactant in aqueous solution. The ECL was assigned to excited J-type PDI dimers. Then, PDI and luminol were used as luminophores to respectively combine with graphite oxide and gold nanoparticles and form potential-resolved ECL nanoprobes. Also, this potential-resolved ECL nanoprobes were respectively functionalized by secondary antibodies (Ab(2)) to construct a low-potential sandwiched ECL immunosensor for tumor markers carcinoembryonic antigen (CEA) and alpha-fetoprotein (AFP) simultaneous determination during linear scanning potential range from -0.6 to 0.6 V. The prepared multiplex immunosensor exhibited sensitive ECL response for CEA at -0.6 V due to PDI and that for AFP at 0.6 V due to luminol, and both linear semilogarithmical ranges were from 0.1 pg to 1 ng mL(-1). In addition, PDI with dual ECL peaks showed enticing prospect of built-in self-calibration for a precise quantitative and bioimaging analysis.

A Proposal for Post Hoc Subgroup Analysis in Support of Regulatory Submission

Summary: In clinical trials, when the primary analysis fails to meet the study objective, the observed positive results in a specific sub-population analysis raise the question of whether they can be used to support regulatory submission. This article proposes several statistical evaluations for confirming the integrity and validity of the observed sub-population analysis results. The evaluations address selection bias, reproducibility, consistency, generalizability, and sensitivity index. A numerical example of a global clinical trial is presented, along with recommendations for incorporating statistical evaluations. Real-world data and evidence are suggested as possible solutions to address regulatory concerns and increase statistical power.

Sequential fully implicit newton method for flow and transport with natural black-oil formulation

Summary: There is an increasing interest in developing robust and efficient sequential methods for reservoir simulation. The sequential fully implicit Newton (SFIN) method addresses the slow sequential coupling convergence issue when flow and transport problems are strongly coupled. However, the original SFIN algorithm requires fixed primary variables during the simulation. In this work, strategies are proposed to handle inconsistent primary variables and extend the SFIN method to the natural black-oil formulation.

A Blockchain and modified Best-worst algorithm enhanced Blackboard model for stranger collaboration based social product design

Integrity verification for scientific papers: The first exploration of the text

Summary: Scientific papers are crucial for academic communication, but many of them lack in-depth research and present core content ambiguously, hindering the progression of science and technology. To address this challenge, the INTEGrity vERification (INTEGER) task is introduced to help researchers assess the integrity of their papers and verify the clarity of each knowledge unit. A multi-task learning model utilizing Tucker decomposition and span-level attention mechanism is proposed to accurately identify terms and their integrity. Experimental results show the effectiveness of the model.
