This study aimed to evaluate the biological activities of two Allium species (garlic and onion) as well as diallyl disulphide (DADS) and dipropyl disulphide (DPDS) as their representative bioactive compounds in a multi-assay experimental design. The genotoxic, antigenotoxic, and lifespan effects of garlic, onion, DADS, and DPDS were checked in Drosophila melanogaster and their cytotoxic, pro-apoptotic, and DNA-clastogenic activities were analyzed using HL60 tumoral cells. All compounds were non-genotoxic and antigenotoxic against H2O2-induced DNA damage with a positive dose-response effect and different inhibition percentages (the highest value: 95% for DADS) at all tested concentrations. Daily intake of Allium vegetables, DADS, or DPDS had no positive effects on flies' lifespan and health span. Garlic and DADS exerted the highest cytotoxic effects in a positive dose-dependent manner. Garlic and DADS exerted a DNA-internucleosomal fragmentation as an index of induced proapoptotic activity on HL60 cells. Allium vegetables and DADS were able to induce clastogenic strand breaks in the DNA of HL60 cells. This study showed the genomic safety of the assayed substances and their protective genetic effects against the hydrogen peroxide genotoxine. Long-term treatments during the whole life of the Drosophila genetic model were beneficial only at low-median concentrations. The chemo-preventive activity of garlic could be associated with its distinctive organosulfur DADS. We suggest that supplementary studies are needed to clarify the cell death pathway against garlic and DADS.

500 years of breeding in the Carthusian Strain of Pura Raza Espanol horse: An evolutional analysis using genealogical and genomic data

Julia Poyato-Bonilla, Nora Laseca, Sebastian Demyda-Peyras, Antonio Molina, Mercedes Valera

Summary: The Carthusian horse, a Pura Raza Espanol strain, has been bred as a closed population for over 500 years. Analysis of its population structure revealed a unique genetic profile with a history of both differentiation and introgression. The study also noted recent reductions in variability and a genetic bottleneck in the population.

Effect of the rob(1;29) translocation on the fertility of beef cattle reared under extensive conditions: A 30-year retrospective study

Jose Maria Jimenez, Rosa Morales, Antonio Molina, Miguel Moreno-Millan, Sebastian Demyda-Peyras

Summary: This study analyzed the impact of rob(1;29) in the Retinta cattle breed and found that cows carrying the heterozygote genotype showed a significant decrease in fertility. After avoiding selection of carrier individuals as breeding bulls for 30 years, the breed's fertility significantly improved and the effect of rob(1;29) on cattle fertility was only significant when the prevalence of carriers was high.

Toxicological and Nutraceutical Screening Assays of Some Artificial Sweeteners

Marcos Mateo-Fernandez, Miguel Josue Gonzalez-Jimenez, Mercedes Del Rio Celestino, Rafel Font, Angeles Alonso-Moraga, Tania Merinas-Amo

Summary: This study evaluated the toxicity of three artificial sweeteners, acesulfame-k, aspartame, and cyclamate. The results demonstrated that these compounds were not toxic or genotoxic in fruit flies, but they decreased the lifespan of the flies. Additionally, aspartame and cyclamate showed protective effects against genotoxins in fruit flies. In cell experiments, acesulfame-k and aspartame exhibited cytotoxicity and the ability to cause DNA damage.

Safety and Protective Activities of Manufactured Alcohol-Free Beers

Tania Merinas-Amo, Mercedes Del Rio Celestino, Rafael Font, Angeles Alonso-Moraga

Summary: There is a growing interest in improving health to achieve better quality of life. Diet plays a crucial role in determining health conditions. In this study, the biological activities of three types of alcohol-free lager beer and one of their most abundant phenols, epicatechin gallate (ECG), were analyzed to identify their potential as nutraceuticals. The study employed Drosophila melanogaster as an animal model and human leukaemia cells (HL-60) for in vitro experiments. The results indicate the safety and protective properties of the tested compounds, with alcohol-free beers and ECG showing promising effects in terms of genotoxic activity, oxidative damage protection, longevity increase, and inhibition of tumour cell growth.

Fine-Scale Analysis of Runs of Homozygosity Islands Affecting Fertility in Mares

Nora Laseca, Antonio Molina, Manuel Ramon, Mercedes Valera, Florencia Azcona, Ana Encina, Sebastian Demyda-Peyras

Summary: The study evaluated the impact of increased inbreeding levels on mare fertility, showing a negative correlation between whole-genome homozygosity and fertility estimated breeding values. The analysis also revealed chromosome-level variability and stronger correlations in a reduced dataset of the 10% most and least fertile individuals. Furthermore, the study identified genomic regions associated with fertility, including candidate genes related to oocyte and embryo development.

Summary: This study conducted a genome-wide association study in horses to identify genomic variants, regions, and candidate genes associated with mare fertility. The results showed potential associations between reproductive efficiency and specific genetic markers, regions, and candidate genes, providing important insights into the genetic mechanisms of mare fertility.

Copy Number Variation (CNV): A New Genomic Insight in Horses

Nora Laseca, Antonio Molina, Mercedes Valera, Alicia Antonini, Sebastian Demyda-Peyras

Summary: This study provides a detailed characterization of CNV regions in the Pura Raza Espanola horse breed, revealing a unique pattern of genomic regions enriched in CNVs and the incidence of CNVs across the entire genome. The study also identifies genes related to olfactory pathways and immune response. The findings contribute to our understanding of CNVs in equines and their impact on genetic and phenotypic variations.

Summary: This study used a genomic approach to investigate the genetic basis of historical Spanish Merino sheep, and found significant genetic differences between them and modern Merino breeds. They also exhibited high variability and richness in terms of genetics, suggesting that they should be protected as a valuable genetic population for improving sheep breeds.

Summary: Chromosomal abnormalities are commonly associated with fertility impairments in horses. This study aimed to analyze the prevalence of chromosomal diseases in a large population of Pura Raza Espanol horses using a two-step molecular tool. Thirteen individuals showed positive results for chromosomal abnormalities, including cases of sex development disorders, blood chimerism, Turner syndrome, Klinefelter syndrome, and complex karyotypes. The overall prevalence in the entire population was about 0.05%, with sex development disorders and blood chimerism occurring at rates of 0.02% and 0.016%, respectively.

Analysis of the genomic landscape of inbreeding in two divergent groups of Spanish Florida goats

Tatiana E. Ziegler, Antonio Molina, Manuel Ramon, Manuel Sanchez, Eva Munoz-Mejias, Alicia Antonini, Sebastian Demyda-Peyras

Summary: The genomic landscape of inbreeding was analyzed in Florida goats using runs of homozygosity (ROH) patterns as a tool to understand phenotypic differences. Inbreeding levels were determined for each individual using ROH-based inbreeding coefficients (F-ROH), and two groups were formed based on these levels. The distribution of ROH was analyzed in each group, showing differences in abundance and distribution among chromosomes. Genomic regions with increased ROH were identified, and candidate genes related to production, fertility, and heat resistance were found within these regions.

Summary: The Bovine Pangenome Consortium (BPC) aims to assemble cattle genomes to create a comprehensive representation of cattle genomic diversity. Their goal is to provide genome assemblies and a community-approved pangenome reference as a public resource to replace breed-specific reference genomes. The BPC invites partners to participate in this collaborative effort and contribute to the development of this community-driven resource. It will serve as a foundation for cattle genomic selection and enable comparison across studies.
