Engineering Metabolism of Chimeric Antigen Receptor (CAR) Cells for Developing Efficient Immunotherapies

Joslyn L. Mangal, Jamie L. Handlos, Arezoo Esrafili, Sahil Inamdar, Sidnee Mcmillian, Mamta Wankhede, Riccardo Gottardi, Abhinav P. Acharya

Summary: This review paper discusses the recent progress in CAR-based therapies for tumors and the challenges faced in treating solid tumors. It emphasizes the advances in CAR T cell therapy for hematological malignancies and the difficulties in applying it to solid tumors. The importance of understanding nutrient and energy supplies in the tumor microenvironment is highlighted, along with the potential of bioengineering technologies in developing metabolically fit CAR-immune cells for cancer treatment.

An in vitro chondro-osteo-vascular triphasic model of the osteochondral complex

Alessandro Pirosa, Riccardo Gottardi, Peter G. Alexander, Dario Puppi, Federica Chiellini, Rocky S. Tuan

Summary: The generation of a novel triphasic model facilitates the study of cellular interactions at the osteochondral tissue interface, promoting differentiation of chondrocytes and osteoblasts. The model utilizes wet-spun poly(ε-caprolactone) and PCL/hydroxyapatite scaffolds in combination with a methacrylated gelatin hydrogel as the polymeric backbone of the constructs.

Drug delivery to the pediatric upper airway

Soheila Ali Akbari Ghavimi, Paul M. Gehret, Matthew R. Aronson, Rossana Schipani, Kyra W. Y. Smith, Ryan C. Borek, John A. Germiller, Ian N. Jacobs, Karen B. Zur, Riccardo Gottardi

Summary: Pediatric upper airway disorders present a challenge due to anatomical complexity and a variety of potential etiologies, with treatment often involving high-dose systemic drugs or invasive surgeries. Innovative airway management technologies designed for adults are limited in pediatric use, but local controlled drug delivery and advancements in drug-eluting technologies show promise in improving outcomes for pediatric upper airway management.

Summary: Articular cartilage is influenced by mechanical loading during development, health, and disease. Researchers have developed a new device called HiT-MACE for high throughput compressive loading of tissue constructs, allowing the investigation of cartilage mechanobiology and the design of effective strategies for degenerative cartilage pathologies.

Three-dimensional and four-dimensional printing in otolaryngology

Irene Chiesa, Carmelo De Maria, Giovanni Vozzi, Riccardo Gottardi

Summary: Each year, thousands of patients face life-threatening ear, nose, and throat disorders, such as tracheal stenosis, or conditions that affect their psychosocial well-being, such as microtia. Tissue engineering offers an exciting alternative by creating bioartificial constructs using three-dimensional scaffolds and human cells that can grow and develop. Bioprinting technologies, which utilize additive manufacturing, are particularly useful in addressing the complex geometries of ear, nose, and throat, allowing for the creation of patient-specific scaffolds with high design flexibility and repeatability.

Amelioration of Subglottic Stenosis by Antimicrobial Peptide Eluting Endotracheal Tubes

Matthew R. Aronson, Amrita Mehta, Ryan M. Friedman, Daniel D. Ghaderi, Ryan C. Borek, Hoang C. B. Nguyen, Kendra S. McDaid, Ian N. Jacobs, Natasha Mirza, Riccardo Gottardi

Summary: We have discovered that modulating the laryngotracheal microbiome can prevent pediatric subglottic stenosis (SGS). In this study, we developed an antimicrobial peptide-eluting endotracheal tube (AMP-ET) that effectively modulates the airway microbiota, resulting in reduced inflammation and resolution of stenosis.
