Structural Diversity and Bioactivities of Peptaibol Compounds From the Longibrachiatum Clade of the Filamentous Fungal Genus Trichoderma

This study examined the structural diversity and bioactivity of peptaibol compounds produced by species from the phylogenetically separated Longibrachiatum Clade of the filamentous fungal genus Trichoderma, which contains several biotechnologically, agriculturally and clinically important species. HPLC-ESI-MS investigations of crude extracts from 17 species of the Longibrachiatum Clade (T. aethiopicum, T. andinense, T. capillare, T. citrinoviride, T. effusum, T. flagellatum, T. ghanense, T. konilangbra, T. longibrachiatum, T. novae-zelandiae, T. pinnatum, T. parareesei, T. pseudokoningii, T. reesei, T. saturnisporum, T. sinensis, and T. orientale) revealed several new and recurrent 20-residue peptaibols related to trichobrachins, paracelsins, suzukacillins, saturnisporins, trichoaureocins, trichocellins, longibrachins, hyporientalins, trichokonins, trilongins, metanicins, trichosporins, gliodeliquescins, alamethicins and hypophellins, as well as eight 19-residue sequences from a new subfamily of peptaibols named brevicelsins. Non-ribosomal peptide synthetase genes were mined from the available genome sequences of the Longibrachiatum Clade. Their annotation and product prediction were performed in silico and revealed full agreement in 11 out of 20 positions regarding the amino acids predicted based on the signature sequences and the detected amino acids incorporated. Molecular dynamics simulations were performed for structural characterization of four selected peptaibol sequences: paracelsins B, H and their 19-residue counterparts brevicelsins I and IV. Loss of position R6 in brevicelsins resulted in smaller helical structures with higher atomic fluctuation for every residue than the structures formed by paracelsins. We observed the formation of highly bent, almost hairpin-like, helical structures throughout the trajectory, along with linear conformation. Bioactivity tests were performed on the purified peptaibol extract of T. reesei on clinically and phytopathologically important filamentous fungi, mammalian cells, and Arabidopsis thaliana seedlings. Porcine kidney cells and boar spermatozoa proved to be sensitive to the purified peptaibol extract. Peptaibol concentrations = 0.3mg ml-1 deterred the growth of A. thaliana. However, negative effects to plants were not detected at concentrations below 0.1 mg ml(-1), which could still inhibit plant pathogenic filamentous fungi, suggesting that those peptaibols reported here may have applications for plant protection.

Identifications of Surfactin-Type Biosurfactants Produced by Bacillus Species Isolated from Rhizosphere of Vegetables

Summary: Surfactins are cyclic lipopeptides consisting of a beta-hydroxy fatty acid and a peptide ring. They have antibacterial, antifungal, and antiviral activities and can be produced by Bacillus species. Bacillus strains isolated from vegetable rhizospheres were identified and tested for surfactin production and inhibition on phytopathogen bacteria and fungi. An HPLC-HESI-MS/MS method was used for quantitative and qualitative analysis, revealing the production of 29 surfactin variants and homologues by more than half of the tested Bacillus strains.

Scale-up of Aflatoxin Purification by Centrifugal Partition Chromatography

Summary: Aflatoxins (AFs) are secondary metabolites that cause diseases in animals and humans. Regulations set concentration limits for AFs in EU food and feed products, requiring pure forms for reference standards. Improvements were made to a liquid-liquid chromatographic method to enhance purification and increase the yield of pure AFs. Through a scale-up process, approximately 2.2 g of AFs could be purified using 8.2 L of solvent in a 250 mL rotor, while 7.8 g of AFs could be prepared using 31 L of solvent in a 1000 mL rotor.

Advantages of Multiplexing Ability of the Orbitrap Mass Analyzer in the Multi-Mycotoxin Analysis

Summary: This study analyzed the antagonistic and defense mechanisms of a soilborne fungus against destructive forest pathogens. The results showed that the antagonistic fungus inhibited the growth of the pathogenic fungus through multiple biocontrol mechanisms, while the pathogenic fungus protected itself from fungal invasion through multiple defense mechanisms. This study provides insights for further exploration of the interaction mechanisms between plant pathogenic fungi and biocontrol agents.

Development of a Multicomponent Microbiological Soil Inoculant and Its Performance in Sweet Potato Cultivation

Summary: The cultivation and consumption of sweet potato are increasing globally, but the use of chemical fertilizers and pesticides may cause pollution. Therefore, there is a need for environmentally-friendly biological solutions. This study developed a microbial inoculant from multiple microorganisms and tested its application in sweet potato cultivation, resulting in increased yield. This is the first successful application of a fungal-bacterial consortium in sweet potato cultivation.

More Than a Diamine Oxidase Inhibitor: L-Aminoguanidine Modulates Polyamine-Related Abiotic Stress Responses of Plants

Summary: L-aminoguanidine (AG) is widely used as an inhibitor in studying plant abiotic stress responses, but its exact mode of action is not well understood. It has been used as a specific diamine oxidase inhibitor and for reducing nitric oxide production. Recent studies suggest its antiglycation effect, but this remains unclear in plants. This review provides a summary of current knowledge about the different targets of AG in plants and recommends using it as a modulator of polyamine-related mechanisms.

Drone-Based Identification and Monitoring of Two Invasive Alien Plant Species in Open Sand Grasslands by Six RGB Vegetation Indices

Summary: Invasive alien species pose a serious threat to biodiversity and ecosystem services and knowing their distribution and spread dynamics is crucial for effective management. Unmanned aerial vehicle (UAV) monitoring, using RGB color-based vegetation indices, was examined for identifying invasive plant species. The TGI and SSI indices were found to be the most suitable for identifying the cover area of common milkweed, while the IF index was the most suitable for blanket flower. However, the methods used were not suitable for determining milkweed shoot and blanket flower inflorescence number due to significant overestimation.

L-Aminoguanidine Induces Imbalance of ROS/RNS Homeostasis and Polyamine Catabolism of Tomato Roots after Short-Term Salt Exposure

Summary: Polyamine catabolism mediated by amine oxidases is important for regulating polyamine homeostasis and related mechanisms during salt stress. The effects of L-aminoguanidine (AG), an inhibitor of copper amine oxidases, on tomato roots exposed to short-term salt stress were studied. AG treatment alleviated the increase in reactive oxygen and nitrogen species caused by salt stress, suggesting its possible antioxidant effect.

Structure-activity correlations for peptaibols obtained from clade Longibrachiatum of Trichoderma: A combined experimental and computational approach

Summary: This study investigated the production of peptaibols by various Trichoderma strains and their correlation with structure-activity relationships. The results showed that T. longibrachiatum f. bissettii SZMC 12546 exerted the lowest minimum inhibitory concentrations (MIC) against Gram-positive bacteria and also inhibited the plant pathogenic Gram-negative Rhizobium radiobacter. Molecular dynamics simulations revealed that the folding dynamics of Group A peptaibols were more restricted and had well-folded helical conformations, which may contribute to their bioactivity against bacteria.
