Estrogen Regulates the Satellite Cell Compartment in Females

Skeletal muscle mass, strength, and regenerative capacity decline with age, with many measures showing a greater deterioration in females around the time estrogen levels decrease at menopause. Here, we show that estrogen deficiency severely compromises the maintenance of muscle stem cells (i.e., satellite cells) as well as impairs self-renewal and differentiation into muscle fibers. Mechanistically, by hormone replacement, use of a selective estrogen-receptor modulator (bazedoxifene), and conditional estrogen receptor knockout, we implicate 17 beta-estradiol and satellite cell expression of estrogen receptor alpha and show that estrogen signaling through this receptor is necessary to prevent apoptosis of satellite cells. Early data from a biopsy study of women who transitioned from peri- to post-menopause are consistent with the loss of satellite cells coincident with the decline in estradiol in humans. Together, these results demonstrate an important role for estrogen in satellite cell maintenance and muscle regeneration in females.

Summary: It has been found that the proliferation and early differentiation of myoblasts are impaired in Marfan syndrome (MFS) mice during muscle regeneration. The fibrotic niche caused by Fbn1 mutations reduces the myogenic potential of satellite cells, affecting structural and functional muscle regeneration. Treatment with the fibrosis inhibitor Ang-(1-7) partially counteracts satellite cell abnormalities and restores myofiber size and contractile force in regenerating muscles.

Summary: This study investigated the associations between early and middle adulthood physical activity (PA) and symptoms of pelvic floor disorders (PFDs) among middle-aged women. The results showed that competitive sports during early adulthood were associated with an increased risk of urge urinary incontinence (UUI) in middle age, while regular PA during early adulthood was associated with an increased risk of fecal incontinence (FI).

In vivo potentiation of muscle torque is enhanced in female mice through estradiol-estrogen receptor signaling

Summary: This study developed a novel method to measure muscle strength in female mice, and found that estradiol enhances posttetanic twitch potentiation (PTP) through the G-protein estrogen receptor (GPER) signaling rather than ERa or ERss signaling pathways. These findings indicate the importance of GPER in mediating the effects of estradiol on muscle strength in female mice.

Proliferation and Maturation: Janus and the Art of Cardiac Tissue Engineering

Summary: During cardiac development and morphogenesis, the differentiation of cardiac progenitor cells into mature cardiomyocytes is regulated by factors that also influence proliferation. Understanding the interaction between proliferation and maturation could enhance the use of human induced pluripotent stem cell-derived cardiomyocytes for modeling adult-level cardiac function in engineered cardiac tissues.

Summary: It is uncertain whether aging before late adulthood and menopause are associated with fat-free mass and fat mass-adjusted resting energy expenditure (REEadj). This study found that there is a significant difference in REEadj between middle-aged and younger women, as well as between middle-aged women with different menopausal statuses. The study also demonstrated that menopause may not contribute to the decline in REEadj in women.

Menopausal symptoms and cardiometabolic risk factors in middle-aged women: A cross-sectional and longitudinal study with 4-year follow-up

Summary: The objective of this study was to examine the associations between menopausal symptoms and cardiometabolic risk factors. The authors conducted a cross-sectional and longitudinal study on a representative population sample of women aged 47-55 years. The results showed modest positive associations between menopausal symptoms and cholesterol and fat mass measures, but these associations disappeared after adjusting for confounders. Menopausal symptoms did not predict changes in risk factors during the follow-up period.
