Single-cell technologies offer an unprecedented opportunity to effectively characterize cellular heterogeneity in health and disease. Nevertheless, visualisation and interpretation of these multidimensional datasets remains a challenge. We present a novel framework, ivis, for dimensionality reduction of single-cell expression data. ivis utilizes a siamese neural network architecture that is trained using a novel triplet loss function. Results on simulated and real datasets demonstrate that ivis preserves global data structures in a low-dimensional space, adds new data points to existing embeddings using a parametric mapping function, and scales linearly to hundreds of thousands of cells. ivis is made publicly available through Python and R interfaces on https://github.com/beringresearch/ivis.

Summary: Surgery is crucial for treating neuroendocrine tumors, with R0 resection being essential. The blood biomarker NETest accurately identifies NETs and elevated scores post-surgery predict recurrence with high accuracy. Surgery decreases NETest levels, but elevated levels post-surgery can indicate residual disease and potential recurrence.

C-type lectin receptor CLEC4A2 promotes tissue adaptation of macrophages and protects against atherosclerosis

Inhye Park, Michael E. Goddard, Jennifer E. Cole, Natacha Zanin, Leo-Pekka Lyytikainen, Terho Lehtimaki, Evangelos Andreakos, Marc Feldmann, Irina Udalova, Ignat Drozdov, Claudia Monaco

Summary: CLEC4A2 is a distinguishing feature of vascular resident macrophages that have athero-protective properties. It promotes monocyte-to-macrophage differentiation and is critical for maintaining vascular homeostasis. CLEC4A2 deficiency leads to dysfunctional cholesterol metabolism and worsens the development of atherosclerosis.

Summary: The developed AI algorithm CovIx can differentiate normal, abnormal, non-COVID-19 pneumonia, and COVID-19 CXRs effectively, achieving an area under the receiver operating characteristic curve for COVID-19 of 0.86. It performs on-par with four board-certified radiologists and is able to triage CXRs rapidly for further patient testing and/or isolation in emergency departments.
