The Effects of Regular Exercise on Circulating Cardiovascular-related MicroRNAs

The purpose of the present study was to examine the effects of regular exercise on the abundance of targeted circulating microRNAs (miRNAs). The present analysis examined 20 previously sedentary adults from the HERITAGE Family Study who completed 20 weeks of endurance exercise training. The expression of 53 miRNAs related to cardiovascular disease were measured in serum collected at baseline and post-training by performing RT-qPCR on the Human Cardiovascular Disease miRNA array (Qiagen, Germany). The effect of regular exercise on circulating miRNAs was assessed using paired t-tests of baseline and post-training expression levels. A false discovery rate threshold of 5% was used to determine significance. Regular exercise resulted in significantly decreased mean serum expression of nine miRNAs (miR-486-5p, let-7b-5p, miR-29c-3p, let-7e-5p, miR-93-5p, miR-7-5p, miR-25-3p, miR-92a-3p, and miR-29b-3p; fold change range: 0.64-83, p =0.0002-0.01) and increased mean expression of five miRNAs (miR-142-3p, miR-221-3p, miR-126-3p, miR-146a-5p, and miR-27b-3p; fold change range: 1.41-3.60, p = 0.001-0.006). Enrichment analysis found that these 14 miRNAs target genes related to over 345 different biological pathways. These results provide further evidence of the effects of regular exercise on the circulating miRNA profile.

Summary: Long and short sleep durations are associated with elevated blood pressure, and genetic factors may play a role in this relationship. Interactions between specific genes and sleep duration were identified, with some genes increasing blood pressure in long sleepers and others increasing blood pressure in short sleepers. This study suggests that the mechanisms regulating blood pressure may be influenced by sleep, providing novel insights into the regulation of sleep-related blood pressure.

Different classes of BMI have varying heritability, with at least 15 gene defects linked to monogenic obesity. Common DNA variants account for most of the BMI heritability, while obesity-promoting alleles have larger effects in those prone to obesity.

Summary: This study conducted a multi-ancestry, genome-wide genetic discovery meta-analysis of lipid levels in approximately 1.65 million individuals, including 350,000 of non-European ancestries. The findings demonstrate that increasing diversity rather than studying additional individuals of European ancestry results in substantial improvements in fine-mapping functional variants and portability of polygenic prediction, with modest gains in the number of discovered loci and ancestry-specific variants. This highlights the importance of including diverse populations in genetic studies to improve the accuracy and equity of polygenic scores in clinical practice.

Summary: Genetic variants have an aggregate effect on baseline and increment in insulin sensitivity index (S-i), focused around cholinergic signalling, calcium, and chemokine signalling. Calcium regulated MEF2A transcription factor is identified as the most statistically significant candidate driving the transcriptional signature associated with increment in insulin sensitivity index.

Summary: This study aimed to assess if considering individual characteristics can make the relationship between heart rate and oxygen uptake reserve during incremental exercise more accurate. The study found that regardless of individual characteristics, the relationship between the percentage of heart rate reserve and oxygen uptake reserve differed from the identity line, and the difference increased after aerobic training.
