The key approach in this work involves the construction of the 2-aminoimidazoline core of nagelamide W from alkene 6 through a cyanamide bromide intermediate. The synthesis of nagelamide W was accomplished with an overall yield of 6.0%.

Summary: Neocryptolepine derivatives, especially 8-Fluoroneocryptolepine (8FNC), have shown promising cytotoxic activity against gastric cancer cells (AGS) with greater selectivity and cytotoxicity compared to commonly used drugs. Structural modifications improved the cytotoxic effect and reduced toxicity to normal gastric cells (GES-1), making 8FNC a potential lead compound for the development of gastric cancer treatment. Mechanistic studies suggest that 8FNC inhibits AGS cell migration and induces apoptosis through the PI3K/AKT signaling pathway.

Summary: In this study, seven new kalihinene diterpenoids, named kalihioxepanes A-G (1-7), were discovered in the sponge Acanthella cavernosa collected from the South China Sea. These compounds possess a rare oxepane ring structure. The structures and configurations were determined through comprehensive spectroscopic analysis, X-ray diffraction, and quantum chemical calculations. Kalihioxepane A (1) exhibited strong cytotoxicity against H69 and K562 tumor cells, while kalihioxepane B (2) showed moderate cytotoxicity against K562 cells.

Among them, lobocatalen A (1) is a new lobane diterpenoid featuring an unusual ether linkage between C-14 and C-18. Additionally, compound 7 exhibited moderate anti-inflammatory activity in zebrafish models and cytotoxic activity against the K562 human cancer cell line.

The structures and absolute configurations of these compounds were determined using various analytical techniques. Among them, sarcoelegan A is composed of a rare tricyclo [11.2.1.0] hexadecane carbon framework, making it the third compound of its kind. Sarcoelegan B and sarcoelegan C possess an unusual seven-membered ether ring, while (+/-)-sarcoelegan D contains a rare peroxo bridge in its seven-membered ring. Furthermore, sarcoelegan A, (+/-)-sarcoelegan D, sarcoelegan E, (+)-sarcoelegan F, and (+)-sarcoelegan H exhibited anti-inflammatory activity in zebrafish, and sarcoelegan C showed anti-thrombotic activity in zebrafish.
