Summary: The permeation enhancer SNAC can form complexes with insulin, improving the oral bioavailability and intestinal mucosal permeability of oral insulin. Microspheres loaded with SNAC/insulin have sustained hypoglycemic activity in diabetic rats. This study provides insight into the interaction between permeation enhancers and payloads and presents a SNAC-based oral insulin delivery system with high oral bioavailability and patient-friendly medication guidance.

Summary: The purpose of this study is to develop and evaluate mucoadhesive in situ nasal gel formulations of loratadine and chlorpheniramine maleate to improve drug bioavailability. Different permeation enhancers were studied for their influence on the nasal absorption of loratadine and chlorpheniramine from in situ nasal gels containing different polymeric combinations. Sodium taurocholate, Pluronic F127, and oleic acid showed significant enhancement in the nasal flux of loratadine, while oleic acid had the most pronounced effect on chlorpheniramine maleate permeation.

Summary: This study presents the process of obtaining chitosan aerogel particles for nasal or inhalation drug delivery systems. The produced chitosan aerogel particles have promising properties for drug delivery, with high surface area, pore volume, and porosity. Experiments demonstrate the effectiveness of chitosan aerogel-clomipramine in targeted drug delivery through the Nose-to-brain mechanism.

Summary: Polysaccharides have been widely used as biomaterials and drugs after oral administration due to their suitable physicochemical properties, good bioactivities and low toxicities. However, studies on their pharmacokinetics and absorption mechanism after oral administration are significantly restricted by the lack of polysaccharide detection methods. With the advancement of polysaccharide detection technologies such as immunoassays, fluorescent and isotopic labelling, the oral pharmacokinetics of polysaccharides have gradually been revealed.

Summary: Medium Chain Triglyceride (MCT) oil was combined with Glyceryl Monostearate (GMS) and Glyceryl Monoolein (GMO) to form stable oleofoams. The addition of 5% GMO resulted in more stable oleogels suitable for pharmaceutical applications. The oleofoams retained their foam characteristics even after 3 months of storage under different temperature conditions.

Summary: Genetic and acquired abnormalities contribute to pancreatic β-cell failure in diabetes. Transcription factors Hnf4a and FoxO1 play important roles through β-cell-specific enhancers. Single gene knockout impairs insulin secretion, but double gene knockout restores β-cell function completely.

Summary: The present study evaluated the effect of gold nanoparticles stabilized by chitosan (CS-AuNPs) and citrate ions (Ci-AuNPs) on skin permeation of sodium fluorescein (NaFI) and rhodamine b base (RhB). CS-AuNPs showed enhanced permeation of NaFI, superior to Ci-AuNPs, and this enhancement was likely due to improved delivery through the transepidermal pathway. However, CS-AuNPs and Ci-AuNPs had no significant effect on the permeability of the lipophilic molecule RhB. CS-AuNPs also exhibited no cytotoxicity towards human skin fibroblast cells.

Summary: In this study, chitooligosaccharide (COS) was identified as a safe, low-cost, and effective oral drug absorption enhancer. The hydrodynamic law of interaction between COS and intestinal mucus layer was revealed, which was associated with absorption benefiting mucus structural reconstruction. A translational strategy was designed to improve the bioavailability of a group of soluble oral drugs by drinking COS solution before administration. This research is also expected to have broader applications, such as reducing drug dosage to avoid gastrointestinal irritation and slowing veterinary antibiotic resistance.

Summary: In this study, chitosan or hyaluronic acid-modified penetration enhancer vesicles were prepared to enhance the skin penetration of silymarin. The vesicles effectively delivered silymarin into the deeper epidermal and dermal layers of the skin, promoting wound healing. Among them, chitosan-modified vesicles showed superior performance compared to hyaluronic acid-modified vesicles.
