Comparative solution and structural studies of half-sandwich rhodium and ruthenium complexes bearing curcumin and acetylacetone

Half-sandwich organometallic complexes of curcumin are extensively investigated as anticancer compounds. Speciation studies were performed to explore the solution stability of curcumin complexes formed with [Rh(eta(5)-C5Me5)(H2O)(3)](2+). Acetylacetone (Hacac), as the simplest beta-diketone ligand bearing (O,O) donor set, was involved for comparison and its Ru(eta(6)-p-cymene), Ru(eta(6)-toluene) complexes were also studied. H-1 NMR, UV-visible and pH-potentiometric titrations revealed a clear trend of stability constants of the acac complexes: Ru(eta(6)-p-cymene) > Ru(eta(6)-toluene) > Rh(eta(5)-C5Me5). Despite this order, the highest extent of complex formation is seen for the Rh(eta(5)-C5Me5) complexes at pH 7.4. Formation constant of [Rh(eta(5)-C5Me5)(H(2)cureumin) (H2O)](+) reveals similar solution stability to that of the acac complex. Additionally, structures of two complexes were determined by X-ray crystallography. The in vitro cytotoxicity of curcumin was not improved by the complexation with these organometallic cations.

Ruthenium(II) polypyridyl complexes with benzothiophene and benzimidazole derivatives: Synthesis, antitumor activity, solution studies and biospeciation

Summary: In this study, two Ru(II)-polypyridyl compounds were prepared and characterized. The solid-state structure was confirmed by X-ray diffraction analysis. The solubility, stability, and lipophilicity of the complexes were investigated under physiological mimetic conditions, as well as their potential uptake by passive diffusion. The complexes exhibited cytotoxicity on ovarian and colon cancer cells and induced late apoptosis and necrosis. They showed weak binding to human serum albumin but significant binding to calf thymus DNA.

Evaluation of In Vitro Distribution and Plasma Protein Binding of Selected Antiviral Drugs (Favipiravir, Molnupiravir and Imatinib) against SARS-CoV-2

Summary: Uncertainties exist in plasma protein binding and blood distribution of the COVID-19 therapeutics favipiravir (FAVI), molnupiravir (MOLNU), and imatinib (IMA). This study investigated the proton dissociation processes, solubility, lipophilicity, and serum protein binding of these drugs. The results showed different degrees of lipophilicity at different pH values. MOLNU was hydrolyzed by blood serum proteins, while FAVI had weak binding to blood proteins and IMA showed high affinity binding to AGP.

Summary: In order to overcome multidrug resistance in cancer chemotherapy, lycorine, a major Amaryllidaceae alkaloid isolated from Pancratium maritimum, was derivatized to obtain 31 new compounds. These compounds showed significant activity in the study of anticancer drug resistance.

Estradiol-Based Salicylaldehyde (Thio)Semicarbazones and Their Copper Complexes with Anticancer, Antibacterial and Antioxidant Activities

Summary: A series of novel estradiol-based salicylaldehyde (thio)semicarbazones and their Cu(II) complexes were synthesized and characterized. The Cu(II)-estradiol-semicarbazone complex structure was determined by X-ray crystallography. The Cu(II) complexes of estradiol-TSC and its N,N-dimethyl derivative showed high cytotoxicity against cancer cells and significant activity against Staphylococcus aureus bacteria strain. Estradiol-TSCs exhibited efficient antioxidant activity, which was decreased by complexation with Cu(II) ions. The exchange of estrone moiety to estradiol did not significantly affect the physicochemical and biological properties.

Isolation of compounds from the roots of Ambrosia artemisiifolia and their effects on human cancer cell lines

Summary: Common ragweed (Ambrosia artemisiifolia L.) is an invasive plant in Europe used in folk medicine. The chemical composition of its above-ground parts is well-studied, but less is known about the metabolites of the roots. In this study, several compounds were isolated from the root extracts, including a new compound called thiophene E (6). Thiophene E exhibited cytotoxic and antiproliferative effects on A-431 epidermoid skin cancer cells. Another compound, taraxerol-3-O-acetate, showed slight antiproliferative effect on gynecological cancer cell lines. These findings highlight the need for further studies on the safety of ragweed root.

Promising anticancer agents based on 8-hydroxyquinoline hydrazone copper(II) complexes

Summary: This study reports the synthesis and characterization of a group of benzoylhydrazones and their Cu(II) complexes. The complexes exhibited moderate-to-strong interaction with bovine serum albumin and showed higher antiproliferative activity compared to the corresponding free ligand and cisplatin. Compound 8 demonstrated the most promising properties with low IC50 values, high induction of oxidative stress and DNA damage, resulting in high rates of apoptosis.

Reversal of Multidrug Resistance by Symmetrical Selenoesters in Colon Adenocarcinoma Cells

Summary: Recently, the anticancer activity of symmetrical selenoesters was investigated in sensitive and resistant human colon adenocarcinoma cells. The compounds showed synergistic interaction with doxorubicin and exhibited ABCB1 inhibitory and apoptosis-inducing effects. Transcriptome analysis revealed that symmetrical selenoesters may influence the redox state of cells and interfere with metastasis formation.

Novel family of [RuCp(N,N)(P)] plus compounds with simultaneous anticancer and antibacterial activity: Biological evaluation and solution chemistry studies

Summary: This article presents the synthesis and properties of a series of novel ruthenium(II)-cyclopentadienyl organometallic compounds. These compounds exhibit high activity in anti-cancer and antibacterial applications. Compounds 6 and 7 show remarkable inhibitory effects on colorectal cancer and non-small cell lung cancer. Additionally, these compounds demonstrate potent antibacterial activity against methicillin-resistant Staphylococcus aureus strains. The study also reveals interactions between these complexes and human serum albumin and calf thymus DNA.

Modulation of the bacterial virulence and resistance by well-known European medicinal herbs

Summary: Salvia officinalis L., Sambucus nigra L., Matricaria chamomilla L., Agrimonia eupatoria L., Fragaria vesca L., and Malva sylvestris L. are plants that have been used in European folk medicine for treating skin wounds and infections. This study investigated their potential as adjuvants for the treatment of antibiotic-resistant bacteria. The results showed that Malva sylvestris had the highest antimicrobial activity, while Fragaria vesca and Sambucus nigra demonstrated potential as efflux pump inhibitors. Overall, these plants have potential for improving antibiotic treatment.

Structure prediction of physiological bis(amino acidato)copper(II) species in aqueous solution: The copper(II) compounds with L-glutamine and L-histidine

Summary: This paper investigates the geometries, energy landscapes, and magnetic parameters of copper(II) amino acid compounds using density functional theory calculations. The results show that certain conformers with histaminate-like and glycine-like modes have low Gibbs free energies and high metal-binding affinities. The predicted conformations reproduce the experimental electron paramagnetic resonance parameters.

Synthesis, characterization and discovery of multiple anticancer mechanisms of dibutyltin complexes based on salen-like ligands

Summary: A series of novel dibutyltin complexes based on salen-like ligands were synthesized and characterized. Complex S03 showed excellent in vitro anticancer activity and induced cancer cell death through multiple mechanisms. This study reveals new mechanisms of organotin complexes and provides new insights into the development of organotin metal complexes as anticancer drugs.

Structural motifs in the early metallation steps of Zn(II) and Cd(II) binding to apo-metallothionein 1a

Summary: Many proteins require a metal cofactor for their function, and these metals play a role in protein folding. The study found that the metallation rates of different metal binding pathways are influenced by pH, with the less cooperative pathway being more affected by low pH conditions. Specific mixtures of structures are formed in the initial steps of metallation, and unfolding the disordered apo-MT structure hinders the formation of these preliminary structures. The compact conformation of the native apo-MT allows for rapid formation of metal-thiolate structures with high affinity, providing protection from oxidation.
