Summary: Signaling by the TGF-beta superfamily is important for neural development and pain processing. In this study, the transcriptional profile of the TGF-beta superfamily members in lumbar DRGs of rats with chronic inflammatory pain was found to be downregulated. Specifically, the signaling of activin/inhibin and BMP/GDF families dramatically decreased. Additionally, peripherally pre-local administration of activins worsened acute inflammatory pain, while activin C improved persistent inflammatory pain by inhibiting astrocyte activation.

Summary: Dysfunctional BMPR2 and eNOS play important roles in the pathogenesis of PAH. Males with PAH usually have a worse prognosis, which may be related to estrogen-associated cardiac and vascular protection. The study showed that BMPR2 mutant female mice were more susceptible to hypoxia-induced PH. The findings suggest that TGF-beta and eNOS contribute to pulmonary microvascular muscularization and sex-linked differences in PAH.

Summary: This study investigated the transcriptional profiles of members of the transforming growth factor-beta superfamily in axotomized dorsal root ganglia. The results showed that the cytokines and signaling pathways of the transforming growth factor-beta superfamily were significantly upregulated after injury. The study also found that activin C can inhibit neuropathic pain through intrathecal injection. These findings provide clues for further understanding the role of the transforming growth factor-beta superfamily in the pathogenesis of neuropathic pain and axon degeneration/regeneration after peripheral nerve injury.

Summary: This study investigated the differences in concentrations of GDF9, BMP15, and the GDF9/BMP15 heterodimer in follicle fluids from women with and without polycystic ovaries (PCO). The results showed that GDF9 levels were significantly higher in women with PCO, while BMP15 levels were significantly lower, resulting in an increased GDF9/BMP15 ratio. This aberrant endocrine environment may serve as a new biomarker for PCO.
