Differential global and MTHFR gene specific methylation patterns in preeclampsia and recurrent miscarriages: A case-control study from North India

Aim: The purpose of the present study is to evaluate and understand the association of global and MTHFR gene specific methylation in preeclampsia and recurrent miscarriages in light of MTHFR C677T polymorphism. Methods: The subjects comprised of recurrent miscarriage cases, their gestation matched controls, preeclampsia cases and matched controls. A set of women at full term were also recruited. Fasting blood sample (similar to 5 ml) was drawn from all the participants followed by DNA extraction, global DNA methylation and MTHFR gene specific methylation. MTHFR C677T polymorphism was analysed by PCR followed by RFLP. Results higher: Global DNA methylation at maternal front (p = 0.04) and hypomethylation of MTHFR gene at fetal front (p = 0.001) might be a characteristic of preeclampsia. Recurrent miscarriage cases were having significantly (p = 0.002) hyper MTHFR gene specific methylation as compared to controls. Women carrying CT genotype were found to be having significantly (p = 0.001) higher global DNA methylation in PE cases and MTHFR gene specific methylation (p = 0.005) in RM cases. Intergenerational analysis revealed similar patterns of global DNA methylation and MTHFR gene specific methylation among both PE and RM cases at maternal and fetal fronts. Conclusion: The study highlights the importance of global DNA methylation in Preeclampsia and MTHFR gene specific methylation in recurrent miscarriages. MTHFR C677T gene polymorphism in association with global and gene specific methylation seem to play a pivotal role in PE and RM respectively.

These results highlight the importance of new dietary guidelines to address the burden of diabetes in South Asia.

Hypoxia Inducible Factors (HIF1 alpha and HIF3 alpha) are differentially methylated in preeclampsia placentae and are associated with birth outcomes

Summary: This study found that DNA methylation and expression of HIF1 alpha and HIF3 alpha were altered in preeclampsia placentae and were associated with preeclampsia, placental weight, and birth outcomes. Specifically, there was significant hypermethylation at cg22891070 of HIF3 alpha in preeclampsia placentae, and marginal hypomethylation at CpG8 in the promoter region of HIF1 alpha in preterm preeclampsia. Furthermore, HIF1 alpha expression was significantly lower in preterm preeclampsia compared to controls. The study also showed a negative correlation between DNA methylation in the HIF1 alpha promoter region and its expression levels, as well as associations between several CpG sites in HIF1 alpha and placental weight and birth outcomes.

Cognitive impairment viz-a-viz genetic and biochemical variations in one carbon metabolic pathway: A population-based study from North India

Summary: This study investigated the association between MTR A2756G, MTRR A66G, and CBS 844ins68 polymorphisms in the one-carbon metabolic pathway (OCMP) and cognitive impairment (CI). The findings showed that MTR A2756G and CBS 844ins68 polymorphisms were not associated with CI, but the AG and AA+AG genotypes of MTRR A66G polymorphism increased the risk of CI. This association became non-significant when vitamin B12 and homocysteine levels were normal. MDR analysis indicated the involvement of the methionine cycle in OCMP in CI.

Genome-wide regulation of Pol II, FACT, and Spt6 occupancies by RSC in Saccharomyces cerevisiae

Summary: RSC is an essential ATP-dependent chromatin remodeling complex in Saccharomyces cerevisiae. It regulates nucleosome-depleted regions (NDRs) by sliding flanking nucleosomes away from NDRs. Depletion of RSC leads to nucleosome encroachment in NDRs and transcription initiation defects. The study compared the effects of catalytic-dead Sth1 and rapid depletion of Sth1 on transcription. Rapid depletion of Sth1 reduces recruitment of TBP and Pol II, while the catalytic-dead mutant exhibits a severe reduction in TBP binding but accumulates Pol II in coding regions. The results suggest a role for RSC in transcription elongation and termination processes.

Age-different BMSCs-derived exosomes accelerate tendon-bone interface healing in rotator cuff tears model
