Summary: Low-grade oncocytic tumour (LOT) of the kidney is a potential novel tumour type with distinct morphological, immunohistochemical, and genetic characteristics. Tumours in this study were found to have alterations in genes related to the tuberous sclerosis/MTOR pathway, as well as PIK3CA. Further research is needed to determine the clinical implications and differences from other renal cancers.

The 2022 revision of the World Health Organization classification of tumors of the urinary system and male genital organs: advances and challenges

Summary: The fifth edition of the WHO classification of urinary and male genital organ tumors, published in 2022, incorporated molecular profiling into the classification of urologic tumors. Major changes include the introduction of molecularly well-defined subtypes of renal tumors and the re-categorization of various tumor types. The new classification also recommends the use of the WHO/ISUP grading for all renal cell carcinomas. Furthermore, there are significant nomenclature changes for different tumor entities. Metastatic, hematolymphoid, mesenchymal, melanocytic, soft tissue, and neuroendocrine tumors are discussed separately in dedicated chapters.

Summary: Optoacoustic imaging enables microscale imaging of endogenous chromophores and has shown potential in identifying superficial microvascular changes associated with human skin pathologies. Raster-scanning optoacoustic mesoscopy has been used to image vascular structures in the gastrointestinal tract in animal models, but accurate localization of intra-abdominal organs is still challenging. This study presents the development of a novel transrectal absorber guide for optoacoustic mesoscopy to enable accurate transabdominal localization and assessment of colonic vascular networks. The potential of this technique is demonstrated through its application in colitis models in mice, highlighting its potential as a monitoring tool for inflammatory bowel diseases.

Summary: In this systematic review and meta-analysis, the cumulative bladder cancer (BC) recurrence rates and efficacy profile of different Bacillus of Calmette-Guerin (BCG) strains were compared. The study found that BCG Tokyo 172 had the lowest recurrence rate among studies with <= 3-year recurrence-free survival (RFS). No significant difference was observed among strains in >3-year RFS outcomes. The genetically different BCG strains RIVM, Tice, and Tokyo 172 showed some potential enhanced benefits.

Vascular Enlargement as a Predictor of Nodal Involvement in Bladder Cancer

Summary: This study aimed to evaluate the presence of perivesical venous ectasia as an indirect sign of lymph node involvement in bladder cancer patients. The results showed that LN short axis > 5 mm, LN enhancement, and Largest Venous Diameter > 3 mm were significantly correlated with positive lymph nodes at pathology. Largest Venous Diameter > 3 mm had high sensitivity and specificity and was an independent predictor.

Summary: Confocal laser endomicroscopy (CLE) is used to examine the architectural differences at the cellular level between mucosa and squamous cell carcinoma (SCC). Analysis of 60 CLE sequences revealed significant differences in cell size and count between SCC cells and healthy epithelial cells, highlighting the importance of these features in identifying SCC.

News in the classification of WHO 2022 bladder tumors

Summary: The fifth edition of WHO Classification of Tumors series for urinary and male genital tract tumors has been published after six years of the fourth edition. New treatment approaches and molecular data on urological cancers have been implemented during these years. Although morphology remains the basis for taxonomy, a molecular approach and new therapeutic modalities like immunotherapy are emerging. More data is required for the application of these advances in routine pathology practice and patient management.
