Over the years, the potential toxicity of anesthetics has raised serious concerns about its safe use during pregnancy. As evidence emerged from research in animal models, showing that some anesthetic drugs are potential teratogenic, the determination of the risk of exposures to anesthetic drugs at early life stages became mandatory. However, due to inaccessibility and ethical constrains related to experimental conditions, the use of early life stages in mammalian models is limited. In this regard, some animal and nonanimal models have been suggested to surpass mammalian use in experimentation. Among them, the zebrafish embryo test has been recognized as a promising alternative in toxicology research, as well as an inexpensive and practical test. Substantial information collected from developmental research following compounds exposure, has contributed to the application of zebrafish assays in research, although only a few studies have focused on the use of early life stages of zebrafish to evaluate the developmental effects of anesthetics. Based on the recent advances of science and technology, there is a clear potential for zebrafish early life stages to provide new insights into anesthetics teratogenicity. This review provides an overview of recent anesthesia research using zebrafish embryos, demonstrating its usefulness to the anesthesia field, discussing the recent findings on various aspects related to the effects of anesthetics during early life development and the strengths and limitations of this model system.

A meta-analytic review of monoterpene for fish anaesthesia

Luis Felix, Raquel Vieira, Sandra M. Monteiro, Carlos Venancio

Summary: The use of anaesthetic agents, including natural substances such as clove oil, as alternatives to the commonly used synthetic anaesthetic MS-222, has been investigated for fish anaesthesia. A meta-analysis of 30 English reports found that 10 different monoterpenes showed high effectiveness in inducing anaesthesia and recovery in fish. The findings also revealed an inverse linear association between anaesthesia induction and recovery time and water bath temperature.

Mitigation of nicotine-induced developmental effects by 24-epibrassinolide in zebrafish

Francisco Pereira, Adriana Pereira, Sandra M. Monteiro, Carlos Venancio, Luis Felix

Summary: This study aimed to evaluate the antioxidant effects of a natural plant hormone, 24-epibrassinolide, against the teratogenic effects of nicotine on zebrafish embryos. The results showed that 24-epibrassinolide reduced nicotine-induced deformities in zebrafish embryos, while improving oxidative stress and metabolic changes. It also alleviated nicotine-induced cognitive dysfunction.

Brassica oleracea Var italica by-Products Prevent Lipid Accumulation and Cell Death in a Liver Cell Model of Lipid Toxicity

Jose P. Castelao-Baptista, Sara A. Valente, Sara Canario, David Oppolzer, Ana Barros, Carlos Venancio, Tania Martins, Luis Antunes, Vilma A. Sardao, Eduardo Rosa, Paulo J. Oliveira

Summary: Obesity, a rising concern in the Eastern world, is often accompanied by non-alcoholic fatty liver disease (NAFLD). Researchers are investigating potential natural interventions to reduce obesity complications. This study focuses on Brassica by-products (BBPs) as an example of edible plant waste and examines the effects of different extracts from broccoli by-products on an in vitro model of fatty acid-induced liver toxicity. The extracts showed a dose-dependent decrease in cell viability of HepG2 cells, but did not compromise mitochondrial function or lipid accumulation. They effectively reduced lipid accumulation induced by fatty acids in HepG2 cells. The extracts' ability to prevent decrease in catalase activity may explain the observed effects.

Anti-Inflammatory, Anti-Oxidative and Anti-Apoptotic Effects of Thymol and 24-Epibrassinolide in Zebrafish Larvae

Germano A. B. Lanzarin, Luis M. Felix, Sandra M. Monteiro, Jorge M. Ferreira, Paula A. Oliveira, Carlos Venancio

Summary: This study investigated the anti-inflammatory, antioxidant, and anti-apoptotic effects of the plant-based products Thymol (THY) and 24-epibrassinolide (24-EPI). Using zebrafish larvae as a model, the researchers found that both compounds reduced neutrophil recruitment, exhibited antioxidant effects by reducing reactive oxygen species (ROS) production, and displayed anti-apoptotic effects. Furthermore, the compounds also decreased the levels of nitric oxide (NO). These findings suggest that THY and 24-EPI have potential as anti-inflammatory and antioxidant agents.
