Summary: This study investigated the bioactivities of pectin oligosaccharides and their regulation effect on gut microbiota. Pectin was partially degraded by acid and enzymatic hydrolysis to obtain two different molecular weight fragments. The solutions of these fragments showed lower viscosity and shear thinning behavior compared to the original pectin oligosaccharides. Further experiments demonstrated that the degraded fragments could relieve constipation symptoms and had better regulation activity on gut microbiota.

Summary: Gut microbiota dysbiosis is a global problem caused by antibiotic overuse. This study found that lentinan, a compound found in mushrooms, has therapeutic effects on antibiotic-driven gut microbial dysbiosis in mice. Lentinan improved the diversity and abundance of the gut microbiota and increased the levels of beneficial bacteria while decreasing harmful bacteria. It also increased the concentrations of short-chain fatty acids and improved colon tissue morphology, reducing inflammation. This study provides a theoretical basis for improving the clinical treatment of broad-spectrum antibiotic side effects.

Summary: The Arctium lappa polysaccharide (ALP) with a molecular weight of 2193 Da was purified and found to be composed mainly of fructose and glucose, with smaller amounts of arabinose, galactose, and mannose. Structural characterization revealed that ALP had a backbone composed of ->(2-beta-D-Fruf-1)n ->. It was shown to have hepatoprotective and antioxidant effects, making it a potential treatment for liver injury.

Summary: Liver disease is a major cause of global mortality, and identifying biomarkers for diagnosing its progression is crucial for improving outcomes. Targeted mass spectrometry is a powerful tool for verifying biomarker candidates and clinical applications, particularly for glycoproteins translation. However, the limitation of analyzing only one sample per run has become apparent. In this study, a high-throughput intact N-glycopeptides quantification strategy was developed, allowing the validation of 20 samples per run with an average analysis time of 3 minutes per sample. The strategy was applied in a cohort of 461 serum samples and identified a panel of 10 IgG N-glycopeptides that have strong clinical utility in evaluating the severity of liver disease.
