Summary: To adapt to stress conditions like infections, cells have developed stress response pathways and innate immune responses, which result in the formation of stress granules (SGs). SGs play a role in antiviral defense, and viruses need to avoid these responses to continue propagating. This study shows that feline calicivirus (FCV) impairs SG assembly by cleaving the protein G3BP1. Additionally, virus-free supernatant from infected cells can induce the formation of SG-like foci, called paracrine granules, which are linked to antiviral activity.

Summary: This study demonstrates that porcine epidemic diarrhea virus (PEDV) infection triggers unfolded protein response (UPR) in Vero E6 cells by activating the PERK/eIF2 alpha pathway, leading to increased expression of proapoptotic protein CHOP and ER oxidoreductase 1 alpha. Inhibition of PERK or CHOP reduces expression of ERO1 alpha and generation of ROS, while inhibition of ERO1 alpha decreases PEDV-induced ROS generation. Genetic or pharmacological inhibition of PERK, CHOP, ERO1 alpha, or ROS suppresses PEDV replication. These findings provide evidence that PEDV manipulates endoplasmic reticulum signaling to disrupt redox homeostasis and promote its replication.

Summary: Avian infectious bronchitis virus (IBV) is a cause of severe production losses in the poultry industry worldwide. This study found that phillygenin, a compound found in Forsythiae Fructus, can inhibit IBV replication by interfering with multiple stages of the virus life cycle. It activates the antiviral response in cells and shows potential as a novel drug for IBV prevention.

Summary: Echinocandin antifungal drugs, including micafungin and its derivatives, showed potent antiviral activities against SARS-CoV-2 and its variants, as well as other viruses. The derivatives Mi-2 and Mi-5 exhibited stronger antiviral activity than micafungin, indicating their potential as antiviral agents. These compounds were found to inhibit intracellular virus replication and retained their activity against drug-resistant SARS-CoV-2 variants. This study provides valuable evidence for the development of antiviral drugs using micafungin derivatives.

Summary: Most drugs used to treat viral disease target specific viruses, but viruses can easily develop resistance to these drugs. Host-targeted antivirals can overcome these limitations. They can be effective against multiple viral pathogens and are especially useful in combating emerging viruses and treating diseases caused by opportunistic agents.

Summary: Acyclovir effectively inhibits viral gene expression, suppresses viral replication, and blocks cytopathic effects and apoptosis induced by CCV in CCO cells. The antiviral activity of acyclovir is not achieved by promoting the expression of immune-related genes, suggesting its potential as a therapeutic agent against CCV.
