miR-21, miR-221 and miR-150 Are Deregulated in Peripheral Blood of Patients with Colorectal Cancer

The Aim of this study was to evaluate the expression levels of miR-21, miR-221, miR-150, let-7a and miR-126a in peripheral blood of 71 patients with colorectal cancer and 80 matched healthy control individuals. We determined expression levels of these microRNAs in peripheral blood samples and used small nucleolar RNA (RNU48) as an internal control. Expression levels of miR-21 (p<0.0001) and miR-221 (p<0.0001) were significantly higher, whereas expression levels of miR-150 (p=0.0054) were significantly lower in the blood samples of patients with colorectal cancer in comparison to the control group. The combination of these three microRNAs enabled us to distinguish patients with colorectal cancer from healthy donors with a sensitivity of 80% and specificity of 74% (p<0.0001). We did not observe any correlation of the studied microRNAs with clinicopathological features of colorectal cancer, indicating that expression of these microRNAs is more likely related to the host response to the tumour than the tumour itself.

Summary: SARS-CoV-2 infections can vary greatly between individuals, with some being asymptomatic while others experience life-threatening disease. While 20% of critical COVID-19 cases are due to inborn errors or autoantibodies, the genetic and immunological factors for resistance to infection itself remain unknown. Studies have shown that autosomal recessive deficiencies in certain genes or enzymes can confer resistance to diseases, suggesting a potential strategy for identifying individuals naturally resistant to SARS-CoV-2 infection.

LncRNA PVT1 is increased in renal cell carcinoma and affects viability and migration in vitro

Summary: This study used next-generation sequencing to comprehensively analyze the expression profiles of lncRNAs in renal cell carcinoma patients, and identified several dysregulated lncRNAs associated with the disease. Among them, PVT1 was found to be significantly dysregulated in renal cell carcinoma and further experiments confirmed its effect on cell viability and migration. These results suggest that PVT1 not only has diagnostic potential, but also may serve as a potential target for the treatment of renal cell carcinoma.

Kinetics of D-lactate and ischemia-modified albumin after abdominal aortic surgery and their ability to predict intestinal ischemia

Summary: This study investigates the perioperative kinetics of D-lactate and ischemia-modified albumin (IMA) and their predictive value in early diagnosis of acute intestinal ischemia. The results suggest that serum levels of IMA and D-lactate can serve as influential predictive markers for postoperative intestinal ischemia, especially after 3 hours from complete declamping of vascular reconstruction.

Two sisters with cardiac-urogenital syndrome secondary to pathogenic splicing variant in the MYRF gene with unaffected parents: A case of gonadal mosaicism?

Summary: Cardiac-urogenital syndrome is a rare syndrome associated with pathogenic variants in the MYRF gene, leading to loss of protein function. It is characterized by complex congenital cardiac defects, pulmonary hypoplasia, diaphragmatic hernia, and urogenital malformations. We report a case of two siblings with extensive developmental defects, who were found to have a novel splicing pathogenic variant in the MYRF gene. The presence of the identical de novo event in siblings suggests gonadal mosaicism, which mimics recessive inheritance. This is the first published case indicating gonadal mosaicism in familial cardiac-urogenital syndrome.

Spinal Metastasis in a Patient with Supratentorial Glioblastoma with Primitive Neuronal Component: A Case Report with Clinical and Molecular Evaluation

Summary: This case report describes a 43-year-old female patient with frontal glioblastoma who developed intraspinal metastasis five months after surgery. Next-generation sequencing analysis revealed a higher number of single nucleotide variants in the metastatic sample compared to the primary tumor. Mutations in NF1, NOTCH3, and ARID1A were identified and may contribute to the invasiveness and metastatic potential of this particular glioblastoma case.
