Micelle Stabilization via Entropic Repulsion: Balance of Force Directionality and Geometric Packing of Subunit

Nanoparticles, 10-30 nm in size, have shown great prospects as nanocarriers for drug delivery. We designed amphiphiles based on 3-helix peptide-PEG conjugate forming 15 nm micelles (defined as 3-helix micelles) with good in vivo stability. Here, we investigated the effect of the site of PEG conjugation on the kinetic stability and showed that the conjugation site affects the PEG chain conformation and the overall molecular architecture of the subunit. Compared to the original design where the PEG chain is located in the middle of the 3-helix bundle, micelle kinetic stability was reduced when the PEG chain was attached near the N-terminus (t(1/2) = 35 h) but was enhanced when the PEG chain was attached near the C-terminus (t(1/2) = 80 h). Quantitative structural and kinetic analysis suggest that the kinetic stability was largely dictated by the combined effects of entropic repulsion associated with PEG chain conformation and the geometric packing of the trimeric subunits. The modular design approach coupled with a variety of well-defined protein stucture and functional polymers will significantly expand the utility of these materials as nanocarriers to meet current demands in nanomedine.

Summary: Lipid nanodiscs formed by mixtures of styrene maleic acid (SMA) copolymers and lipid membranes are important tools for studying membrane proteins, with the formation pathways depending strongly on factors such as temperature, lipid/ copolymer ratio, and lipid type. The initial aggregation of copolymer in solution leads to insertion of its styrene units into the lipid bilayer hydrocarbon region, causing fractures in the membrane.

Molecular Transport and Growth of Lipid Vesicles Exposed to Antimicrobial Peptides

Summary: This study investigates the effect of adding the antimicrobial peptide indolicidin on lipid transport, vesicular growth, and lipid exchange rates in lipid vesicles. The results show that the peptide accelerates lipid transport and induces limited vesicular growth, with DMPG exhibiting faster kinetics. Vesicular growth is mainly associated with peptide-mediated lipid reorganization, which slightly affects overall exchange kinetics.

Transition kinetics of mixed lipid:photosurfactant assemblies studied by time-resolved small angle X-ray scattering

Summary: Photoswitchable surfactants are used in the design of light-responsive colloids and self-assemblies. This study investigates the intermediate lipid-surfactant assemblies formed during fast isothermal photo-triggered transitions. The experiments reveal that the transient assemblies detected during the photo-triggered kinetics are similar to the stationary states. Additionally, the reorganization speed of surfactant-enriched assemblies is faster than that of lipid-enriched ones.

A trimeric coiled-coil motif binds bacterial lipopolysaccharides with picomolar affinity

Summary: Alpha-helical coiled-coils are common protein structures found in all living organisms. Modified coiled-coil sequences have been widely used in biotechnology, vaccine development, and biochemical research to induce protein oligomerization and form self-assembled protein scaffolds. In this study, a trimeric variant of a peptide derived from the yeast transcription factor GCN4, called GCN4-pII, was shown to bind bacterial lipopolysaccharides (LPS) with high affinity. LPS molecules are toxic glycolipids found in the outer membrane of Gram-negative bacteria. The researchers also demonstrated the ability of GCN4-pII to break down LPS micelles in solution. These findings suggest that GCN4-pII and its derivatives could be used for novel LPS detection and removal solutions with important applications in the production and quality control of biopharmaceuticals and other biomedical products, where even trace amounts of residual LPS can be deadly.

Beyond the standard model of solubilization: Non-ionic surfactants induce collapse of lipid vesicles into rippled bilamellar nanodiscs

Summary: This study investigates the structural pathways and metastable structures involved in the solubilization of lipid membranes using the non-ionic detergent Triton X-100. The findings reveal a unique ripple phase of ordered rippled bilamellar disc structures formed when moderate amounts of detergent are added to lipid vesicles, suggesting a combination of water structure destabilization and osmotic pressure causing interbilayer attraction. The study highlights the diversity in assembly pathways of simple lipids and detergents and their implications for membrane solubilization.
