The Role of Fibroblast Growth Factor-18 in Follicular Atresia in Cattle

Although the various members of the fibroblast growth factor (FGF) family are generally mitotic, one member, FGF18, has been shown to increase the rate of apoptosis of ovarian granulosa cells. In the present study, we first determined whether granulosa cells express FGF18 and we then explored the mechanism through which FGF18 increases apoptosis in vitro. Under culture conditions that favored estradiol secretion and CYP19A1 expression, granulosa FGF18 mRNA levels were barely detectable; however, withdrawing gonadotropic support (follicle-stimulating hormone or insulin-like growth factor 1) reduced levels of CYP19A1 mRNA and increased abundance of mRNA encoding the death ligand FASLG and FGF18. Addition of FGF18, but not FGF2, FGF10, or EGF, increased the proportion of apoptotic cells and frequency of caspase 3 activation, and these effects were abrogated by coculture with estradiol. Addition of FGF18 decreased abundance of mRNA encoding the antiapoptotic proteins GADD45B and MDM2, and increased that encoding the proapoptotic protein BBC3; these effects were reversed by coculture with estradiol. The physiological relevance of FGF18 was determined using an in vivo model: injection of FGF18 directly into growing bovine dominant follicles caused cessation of follicle growth by 24 h after injection. Collectively, these data demonstrate that FGF18 is proapoptotic in vivo and may act through a mechanism involving the BBC3-MDM2 pathway.

The Hippo Pathway Effectors YAP and TAZ Regulate LH Release by Pituitary Gonadotrope Cells in Mice

Summary: The study shows that YAP and TAZ play critical roles in gonadotropin synthesis and secretion. Deletion of Yap and Taz genes resulted in increased levels of FSH and LH in male mice, as well as increased testosterone levels and testis weight. Female mice had increased levels of LH and exhibited a hyperfertility phenotype. Loss of YAP/TAZ did not affect the expression of gonadotropin subunit genes, but increased LH secretion. Furthermore, pharmacologic inhibition of YAP binding to TEAD transcription factors increased LH secretion without affecting Lhb gene expression. The study suggests that YAP/TAZ may regulate LH secretion by modulating the expression of secretory granule cargo proteins.

Summary: This study found that the inhibition of the interaction between the Hippo effector YAP or its transcriptional co-activator TAZ with the TEAD family of transcription factors is critical for the cumulus expansion-related events induced by the EGF network in cumulus-oocyte complexes (COCs). These findings provide important insights into the functional relevance of cumulus expansion and oocyte maturation in monovulatory species, and may serve as potential targets to improve the efficiency of in vitro maturation (IVM) systems.

Expression profile of key genes involved in DNA repair mechanisms in bovine cumulus cells cultured with bovine serum albumin or fetal calf serum

Summary: This study characterized the transcript expression profile of genes involved in DNA repair mechanisms in bovine cumulus cells cultured in serum-free medium. The results showed higher mRNA expression of TP53BP1 and BRCA1 genes and lower mRNA expression of TNFAIP6 gene in cumulus cells cultured with BSA. This suggests increased apoptosis rate and DNA damage, which may explain the reduced developmental potential of bovine oocytes in serum-free medium.

Effect of Inactivation of Mst1 and Mst2 in the Mouse Adrenal Cortex

Summary: Recent research has shown that the Hippo signaling pathway is crucial for the development and maintenance of the adrenal cortex. The upstream kinases MST1 and MST2 are expressed in the mouse adrenal cortex, with MST2 specifically expressed in the zona glomerulosa. Conditional deletion of Mst1/2 in steroidogenic cells led to the accumulation of adrenal gonadal primordium-like progenitor cells, without affecting hormone secretion. Gene expression analysis revealed overexpression of downstream target genes of YAP, the main effector of Hippo signaling, and underexpression of genes normally specific to or overexpressed in the zona glomerulosa.

Equine Lacrimal Gland Adenocarcinoma

Summary: This study reports a rare case of equine lacrimal gland adenocarcinoma and its treatment by surgical excision and intralesional chemotherapy. The efficacy of this treatment method was relatively effective. The study highlights the importance of accurate diagnosis and differentiation of lacrimal gland adenocarcinoma from other orbital neoplasms in horses.
