Summary: The upregulation of DDX5 (p68) is associated with various cancers, especially colon adenocarcinomas. MGMT has been identified as a major contributor to chemoresistance. This study reveals that p68 is a transcriptional co-activator of the MGMT gene and enhances its transcription through interactions with beta-catenin, RelA, and SP1 transcription factors.

Summary: Recent research reveals that the base excision repair and MGMT-mediated DNA repair mechanisms can interact with each other in a non-catalytic manner, enhancing the repair of O(6)meG. This interaction is mainly achieved through PARylation by PARP1. In addition, targeting PARP1 and MGMT inhibition could be a more effective cancer therapy.

Up-Regulation of Cyclooxygenase-2 (COX-2) Expression by Temozolomide (TMZ) in Human Glioblastoma (GBM) Cell Lines

Summary: TMZ-resistance is a major limitation in the treatment of glioblastoma (GBM). COX-2/PGE2 system plays a role in GBM TMZ resistance, with a dose-dependent up-regulation of COX-2 expression observed in TMZ-treated cells. Combination of NS398 with TMZ can reduce cell proliferation, induce cell cycle arrest and apoptosis, and prevent the up-regulation of COX-2, beta-catenin, MGMT, and SOX-2 in GBM cells.

Mismatch repair proteins play a role in ATR activation upon temozolomide treatment in MGMT-methylated glioblastoma

Summary: Bromodomain and extra-terminal tail (BET) proteins are potential epigenetic targets in cancer, including glioblastoma. BET inhibitors disrupt the histone code-gene transcription link. BET-induced differential gene expression in glioblastoma derived-spheres revealed 6 distinct response patterns. The O-6-methylguanine-DNA methyltransferase (MGMT) gene, a known resistance factor for glioblastoma treatment, showed consistent downregulation in response to BET inhibitors.

Summary: The aim of this study was to evaluate the clinical efficacy of adding temozolomide to preoperative capecitabine-based chemoradiotherapy in patients with locally advanced rectal cancer, and to validate O-6-methylguanine DNA methyltransferase methylation status as a predictive marker for temozolomide combined regimens. The results showed that the addition of temozolomide tended to improve pathological complete response rates in locally advanced rectal cancer patients, particularly in those with methylated O-6-methylguanine DNA methyltransferase status. Further investigation is needed to determine the predictive role of O-6-methylguanine DNA methyltransferase status as a biomarker for chemotherapeutic agents and radiotherapy.
