ARHGAP17 suppresses tumor progression and up-regulates P21 and P27 expression via inhibiting PI3K/AKT signaling pathway in cervical cancer

ARHGAP17 has long been thought to be involved in the maintenance of tight junction and epithelial barrier. Recently, a few Rho GTPase activating proteins (RhoGAPs) have been identified as tumor suppressors in some human cancers. The present study aimed to explore ARHGAP17 expression in cervical cancer and the possible function in tumor progression. ARHGAP17 expression in cervical cancer cell lines was assessed by RT-PCR and Western blotting. ARHGAP17 expression in cervical cancer tissues and normal tissues was assessed by immunohistochemistry. The cell proliferation was determined using CCK-8 in vitro and subcutaneous xenograft model in vivo. Here, we showed lower expression of ARHGAP17 in cell lines and human cervical cancer samples. Manipulation of ARHGAP17 affected cell proliferation in vitro and tumor growth in vivo. Furthermore, the phosphorylation of AKT was enhanced in ARHGAP17 silencing cervical cancer cells. ARHGAP17 can elevate P21 and P27 expression level through inhibiting PI3K/AKT signaling pathway. Stepwise investigations demonstrated that ARHGAP17 suppressed malignant phenotype of cervical cancer cells via inhibiting PI3K/AKT signaling pathway. These results reveal that ARHGAP17 functions as a tumor suppressor in cervical cancer that suppresses tumor growth, at least partly, through inhibition of PI3K/AKT signaling and up-regulation of P21 and P27 expression.

Identification of differentially expressed proteins and clinicopathological significance of HMGB2 in cervical cancer

Summary: Using iTRAQ-based mass spectrometry analysis, HMGB2 was found to be significantly up-regulated in cervical cancer tissues compared to high-grade squamous intraepithelial lesion tissues and normal cervical tissues. High HMGB2 expression was associated with primary tumor size, invasion, and tumor stage, suggesting HMGB2 as a potential prognostic biomarker and therapeutic target for cervical cancer.

Vaginal Microbiome Dysbiosis is Associated with the Different Cervical Disease Status

Summary: The composition of the vaginal microbiome is associated with cervical disease, particularly cervical cancer. This study found that higher microbiome diversity and depletion of Lactobacillus species, especially L. crispatus, were associated with the severity of cervical disease. High-risk HPV16 infection also correlated with higher microbiome diversity and depletion of Lactobacillus in high-grade cervical diseases. The presence of specific bacteria in the vaginal microbiome, such as Fannyhessea vaginae, Prevotella, Bacteroides, Finegoldia, Vibrio, Veillonella, Peptostreptococcus, and Dialister, characterized the CC group.

Altered vaginal eukaryotic virome is associated with different cervical disease status

Summary: In this pilot exploratory study, the vaginal eukaryotic virome in women with different levels of cervical lesions was investigated using viral metagenomics. An altered eukaryotic virome was observed in women with different levels of lesions and Lacto-bacillus profiles. Anelloviruses and papillomaviruses were the most commonly detected eukaryotic viruses of the vaginal virome, and their abundance and diversity were associated with low-grade squamous intraepithelial lesion (LSIL) and cervical cancer (CC).

Genome-wide regulation of Pol II, FACT, and Spt6 occupancies by RSC in Saccharomyces cerevisiae

Summary: RSC is an essential ATP-dependent chromatin remodeling complex in Saccharomyces cerevisiae. It regulates nucleosome-depleted regions (NDRs) by sliding flanking nucleosomes away from NDRs. Depletion of RSC leads to nucleosome encroachment in NDRs and transcription initiation defects. The study compared the effects of catalytic-dead Sth1 and rapid depletion of Sth1 on transcription. Rapid depletion of Sth1 reduces recruitment of TBP and Pol II, while the catalytic-dead mutant exhibits a severe reduction in TBP binding but accumulates Pol II in coding regions. The results suggest a role for RSC in transcription elongation and termination processes.

miR-124-3p regulates the involvement of Ptpn1 in testicular development and spermatogenesis in mouse

Age-different BMSCs-derived exosomes accelerate tendon-bone interface healing in rotator cuff tears model

Summary: This study found that exosomes secreted by young BMSCs can promote the healing of tendon-bone interface after rotator cuff tears. These exosomes can improve extracellular matrix remodeling, osteogenic differentiation, angiogenesis, and stemness of tendon-derived stem cells (TDSCs). The exosomes from young BMSCs have better effects compared to those from aged BMSCs.
