Summary: A new platform was used to genotype HPV DNA in cancer tissues of 361 gastric cancer (GC) and 89 oropharyngeal squamous cell carcinoma (OPSCC). The results showed that 10 GC and 2 OPSCC were infected with HPV16, suggesting a possible involvement of high-risk HPV in gastric carcinoma.

Summary: Infection with oncogenic strains of HPV can lead to malignant progression and tumorigenesis. In this study, a novel thermostable enzyme chemistries-based assay was developed for rapid clinical HPV screening. The use of thermostable enzymes provided faster cHPV-DNA extraction and amplification, resulting in improved overall assay time and achieving excellent sensitivity and specificity in human plasma specimens.

Summary: Human papillomavirus (HPV) is considered the main cause of increasing incidence rates of oropharyngeal squamous cell carcinoma (OPSCC), with global burden predicted to exceed that of cervical cancer. Different molecular profiles for HPV-related OPSCC offer new targeted therapies and immunotherapy approaches. Epigenetic and microbiome-based biomarkers show promise for OPC screening and treatment.

Summary: Clinical trial data and real-world evidence suggest that the AS04-adjuvanted vaccine targeting human papillomavirus types 16 and 18 provides nearly 90% protection against cervical intraepithelial neoplasia grade 3 or higher among women vaccinated before sexual debut. This high efficacy is not fully explained by cross-protection, and although the vaccine does not affect clearance of prevalent infections, it may accelerate clearance of newly acquired infections. However, pooled data from 2 large-scale randomized controlled trials did not show a significant effect in expediting clearance of incident infections.

Summary: Human papillomavirus (HPV) is associated with a variety of cancers and vaccination has been effective in preventing infections. However, the burden of HPV-related cancers remains high worldwide. Circulating HPV-induced cancer DNA is being studied as a potential biomarker for optimizing cancer control.

Summary: HPV testing is mandatory for newly diagnosed OPSCC in order to assist in treatment decisions and prognosis. FNA is commonly used for OPSCC but can yield indeterminate results for HPV testing. In this study, we propose the use of ctHPVDNA to determine HPV status in cases with indeterminate FNA results. We report three cases where ctHPVDNA successfully confirmed HPV status. If validated, this non-invasive assay could eliminate the need for repeat FNAs or biopsies solely for determining HPV status.

Summary: The aim of this study was to assess and compare the nutritional status of patients with HPV-related and non-HPV-related oropharyngeal cancer before and after radiotherapy or chemoradiotherapy. The study found that regardless of HPV status, patients with OPC can develop malnutrition during RT/CRT. Higher BMI was associated with better overall survival and disease-free survival in the HPV- group, while higher total lymphocyte count (TLC) was associated with better disease-free survival in the HPV+ group.

Summary: This study identified significantly differentially expressed circulating miRNAs in plasma samples of GBM patients, suggesting their potential as diagnostic biomarkers. Functional annotation and pathway enrichment analysis of target genes of these miRNAs were conducted to assess their functional connections. Plasma microRNA profiles show promise as diagnostic and prognostic biomarkers for GBM, with further validation studies needed.

Summary: High-throughput screening of FDA-approved drugs identified BET inhibitor OTX015 as a top suppressor of ESR1 mutant cell growth, showing superior efficacy compared to fulvestrant. When combined with CDK4/6 inhibitor abemaciclib, OTX015 induced more potent tumor regression than current standard-of-care treatment.

Summary: Over 70% of oropharyngeal head and neck squamous cell carcinoma (HNSC) cases in the United States are positive for human papillomavirus (HPV). This study identified robust expression of Synaptogyrin-3 (SYNGR3) gene in immune cells of HPV(+) squamous cancers and demonstrated that codetection of SYNGR3 and p16 by immunohistochemistry (IHC) can more reliably identify the low-risk subgroup of patients that may be appropriate for deescalation treatments.
