GLP-1's role in neuroprotection: a systematic review

Glucagon-like peptide 1 (GLP-1) is a target for treatment of diabetes; however, its function in the brain is not well studied. In this systematic review, we aimed to analyze the neuroprotective role of GLP-1 and its defined mechanisms.Methods: We searched Web of Science' and Pubmed' to identify relevant studies using GLP-1 as the keyword. Two hundred and eighty-nine clinical and preclinical studies have been included. Data have been presented by grouping neurodegenerative, neurovascular and specific cell culture models.Results: Recent literature shows that GLP-1 and its agonists, DPP-4 inhibitors and combined GLP-1/GIP molecules are effective in partially or fully reversing the effects of neurotoxic compounds, neurovascular complications of diabetes, neuropathological changes related with Alzheimer's disease, Parkinson's disease or vascular occlusion. Possible mechanisms that provide neuroprotection are enhancing the viability of the neurons and restoring neurite outgrowth by increased neurotrophic factors, increasing subventricular zone progenitor cells, decreasing apoptosis, decreasing the level of pro-inflammatory factors, and strengthening blood-brain barrier.Conclusion: Based on the preclinical studies, GLP-1 modifying agents are promising targets for neuroprotection. On the other hand, the number of clinical studies that investigate GLP-1 as a treatment is low and further clinical trials are needed for a benchside to bedside translation of recent findings.

More translational research is required to further evaluate its impact on human reward system related disorders.

Generation of TRAIL-resistant cell line models reveals distinct adaptive mechanisms for acquired resistance and re-sensitization

Summary: The study identified acquired TRAIL resistance models in glioblastoma cells and showed that proteasome inhibitor Bortezomib can effectively overcome TRAIL resistance by upregulating death receptors and pro-apoptotic proteins. Inhibition of the NF-kappa B pathway increased sensitivity to TRAIL in the most resistant cells. The findings suggest the critical importance of combining proteasome inhibitors and pro-apoptotic ligands to overcome acquired resistance.

Tumor Cell Infiltration into the Brain in Glioblastoma: From Mechanisms to Clinical Perspectives

Summary: Glioblastoma is an extremely aggressive and invasive primary brain tumor with poor prognosis. Understanding the molecular mechanisms of glioblastoma invasion and developing new therapeutic approaches are of utmost importance. Efforts have been made to uncover the routes followed by glioblastoma cells, their interaction with the brain environment and extracellular matrix, and the role of cell adhesion and extracellular matrix remodeling. However, there is currently no approved therapy targeting the invasive phenotype.

Summary: By applying fractionated radiotherapy over a long time and selecting irradiation-survivor glioblastoma cells, clinically relevant irradiation-exposed models were generated. The cells exhibited slower growth, resistance to secondary irradiation, upregulation of DDR-related genes, and better DNA repair capacity. The utility of combining DDR inhibitors and irradiation was demonstrated for both naive and IR-Surv glioblastoma cells, as long as cells are refrained from hypoxic conditions.

Clinical Validation of SERS Metasurface SARS-CoV-2 Biosensor

Summary: In this study, a surface-enhanced Raman spectroscopy (SERS) metasurface optimized with genetic algorithm (GA) was developed to detect SARS-CoV-2 directly using unprocessed saliva samples. After validation of system performance, the SERS substrate was tested with inactivated SARS-CoV-2 virus for virus detection, viral load analysis, cross-reactivity, and variant detection using machine learning models. With 36 PCR positive and 33 negative clinical samples, SARS-CoV-2 positive samples were detected from Raman spectra with 95.2% sensitivity and specificity.
