We present software to characterize and rank potential therapeutic (drug) targets with data from public databases and present it in a user-friendly format. By understanding potential obstacles to drug development through the gathering and understanding of this information, combined with robust approaches to target validation to generate therapeutic hypotheses, this approach may provide high quality targets, leading the process of drug development to become more efficient and cost-effective.

A polygenic resilience score moderates the genetic risk for schizophrenia

Jonathan L. Hess, Daniel S. Tylee, Manuel Mattheisen, Anders D. Borglum, Thomas D. Als, Jakob Grove, Thomas Werge, Preben Bo Mortensen, Ole Mors, Merete Nordentoft, David M. Hougaard, Jonas Byberg-Grauholm, Marie Baekvad-Hansen, Tiffany A. Greenwood, Ming T. Tsuang, David Curtis, Stacy Steinberg, Engilbert Sigurdsson, Hreinn Stefansson, Kari Stefansson, Howard J. Edenberg, Peter Holmans, Stephen Faraone, Stephen J. Glatt

Summary: This study proposed a theoretical framework based on genetic variations that promote resilience to highly heritable polygenic disorders, such as schizophrenia. By establishing a procedure to identify unaffected individuals with relatively high polygenic risk for the disease and generating the first known polygenic resilience score, this work laid a foundation for finding resilience variants for any complex, heritable disorder.

Summary: The study identified differentially methylated positions (DMPs) in blood DNA methylation profiles associated with psychosis, schizophrenia, and treatment-resistant schizophrenia. Analysis across datasets revealed DMPs with evidence of colocalization to regions nominated by genetic association studies of the diseases. The findings suggest that DNA methylation data can serve as molecular biomarkers for treatment-resistant schizophrenia and reveal environmental factors associated with psychosis.

Summary: The study found significant sex-dependent genetic risk in schizophrenia, major depressive disorder, and bipolar disorder, with implications for genes related to neuronal development, immune functions, and vascular functions across and within these disorders. This suggests substantial genetic overlap between sexes in mood and psychotic disorders, with sex-specific effects enriched for genes involved in various physiological functions.

A human tau seeded neuronal cell model recapitulates molecular responses associated with Alzheimer's disease

Elena Ficulle, Sarubini Kananathan, David Airey, Severine Gharbi, Neil Humphryes-Kirilov, James Scherschel, Charlotte Dunbar, Brian J. Eastwood, Emma Laing, David A. Collier, Suchira Bose

Summary: Seeding rat neurons with tau isolated from Alzheimer's disease brains can induce an increase in endogenous tau inclusions. Transcriptomic analysis identified 1075 differentially expressed genes in these cells, enriched for lipid/steroid metabolism and neuronal/glial cell development. Several genes were found to be correlated with tau inclusion formation at both transcriptomic and proteomic levels.
